A poly(ethylene glycol) three-dimensional bone marrow hydrogel

Lauren E. E. Jansen et al.

Summary: Three-dimensional hydrogels made from synthetic polymers can mimic the extracellular matrix (ECM) of real tissues, but they have limitations in terms of the diversity and specificity of bioactive peptides. To overcome these limitations, researchers have developed a bone marrow hydrogel that incorporates the complex and specific protein signature of bone marrow. This tissue-centric approach allows for better replication of the elasticity and structure of bone marrow, and provides a wide range of bone marrow-specific and cell-instructive peptides.

Mimicking Bone Extracellular Matrix: From BMP-2-Derived Sequences to Osteogenic-Multifunctional Coatings

Lluis Oliver-Cervello et al.

Summary: Cell-material interactions can be regulated by mimicking bone extracellular matrix on the surface of biomaterials. Synthetic osteogenic domains derived from BMP-2 are gaining attention as a strategy for bone regeneration without clinical risks associated with the molecule. Screening of the BMP-2 epitopes identified peptides that enhance cell adhesion and osteogenic differentiation. The peptides were used to functionalize titanium surfaces, showing improved bone formation and reduced fibrous tissue thickness.

Enhanced bone regeneration via spatiotemporal and controlled delivery of a genetically engineered BMP-2 in a composite Hydrogel

Xin Chen et al.

Summary: The modification strategy using His6-T4L enhances the stability and solubility of BMP-2, allowing for controlled release and maximizing its activity to enhance bone regeneration. Additionally, the gelatin-based hydrogel incorporated with MPs-His6-T4L-BMP2 displays favorable properties for bone defect repair.
