Three-dimensional in vitro models of neuromuscular tissue

Paolo Raffa et al.

Summary: Skeletal muscle is reliant on the clear three-dimensional organization of myofibers and communication between myofibers and the nervous system for development and function. Various pathologies affecting muscle function are linked to alterations in the interaction between motor neurons and myofibers. Recent research indicates that three-dimensionality and integration of multiple cell types are essential for unveiling pathophysiologically relevant phenotypes in skeletal muscle modeling.

Remodeling and Fibrosis of the Cardiac Muscle in the Course of Obesity-Pathogenesis and Involvement of the Extracellular Matrix

Jagoda Kruszewska et al.

Summary: Obesity is a growing problem, affecting two-thirds of adults and increasing the risk of cardiovascular diseases. Chronic obesity can also lead to heart failure with preserved ejection fraction. Excessive collagen deposition in the extracellular matrix during obesity can stiffen the myocardium and impair heart function. This review discusses the rationale and potential factors behind this process.

Intrinsic Differences in Spatiotemporal Organization and Stromal Cell Interactions Between Isogenic Lung Cancer Cells of Epithelial and Mesenchymal Phenotypes Revealed by High-Dimensional Single-Cell Analysis of Heterotypic 3D Spheroid Models

Maria L. Lotsberg et al.

Summary: The lack of adequate preclinical models hinders cancer drug development, and the heterotypic multicellular spheroids offer a promising solution. This study extensively characterizes the spheroids and demonstrates their applicability in evaluating the impact of cancer cell phenotype in drug screening experiments. High-dimensional and spatially resolved imaging mass cytometry analysis provides detailed insights into the heterogeneity of cancer-stroma interactions and epithelial phenotypic plasticity, which are crucial factors in therapy resistance in non-small cell lung cancer (NSCLC).

Super Liquid-repellent Surfaces and 3D Spheroids Growth

Michele Ferrari et al.

Summary: The composition and surface features of materials play a crucial role in controlling cell adhesion to surfaces, which significantly impact various aspects of cell functions in biomedical applications. Recent studies have shown that culturing 3D cell aggregates on low wettable surfaces can provide reliable conditions for studying tumor and non-tumor cell lines. Promoting cell aggregate formation through highly hydrophobic surfaces is particularly important for spheroid formation.

Matrigel 3D bioprinting of contractile human skeletal muscle models recapitulating exercise and pharmacological responses

Angela Alave Reyes-Furrer et al.

Summary: The researchers adapted a 3D bioprinting platform for microvalve-based drop-on-demand printing of cell-laden Matrigel containing primary human muscle precursor cells, which could aid in the development of drugs against muscle wasting diseases. This new method allows for automated engineering of 3D human skeletal muscle models in microplates and functional compound screening, addressing the lack of medication for muscle wasting diseases.

Regulation of Skeletal Muscle Satellite Cell Differentiation by Omega-3 Polyunsaturated Fatty Acids: A Critical Review

Peter O. Isesele et al.

Summary: Skeletal muscle is primarily composed of myofibers, with their size and muscle mass being influenced by various pathological and physiological conditions. Satellite cells play a crucial role in muscle regeneration, but a reduced number and impaired myogenic function may lead to muscle disorders. The function of satellite cells is affected by mitochondrial function, inflammation, and Omega-3 PUFAs.
