A Green Synthesis of Carbene-Metal-Amides (CMAs) and Carboline-Derived CMAs with Potent in vitro and ex vivo Anticancer Activity

Summary: The modularity and ease of synthesis of carbene-metal-amide (CMA) complexes based on the coinage metals (Au, Ag, Cu) and N-heterocyclic carbenes (NHCs) as ancillary ligands offer potential applications in various fields beyond photochemistry and catalysis. This study presents an improved synthesis method for these compounds, eliminating the use of toxic solvents, and expands their scope by incorporating carbolines as amido fragments. The newly synthesized complexes exhibit excellent cytotoxicity against cancer cell lines and high-grade serous ovarian cancer (HGSOC) tumoroids, showing promise for future development.

The Genomic Landscape of Early-Stage Ovarian High-Grade Serous Carcinoma

Summary: Late-stage and early-stage high-grade serous carcinomas (HGSCs) of the ovary exhibit highly similar patterns of mutation and copy-number alterations. However, late-stage HGSCs show distinct copy-number signature exposures consistent with whole-genome duplication. Further analysis is needed to determine whether these differences reflect genuine biological disparities between early-stage and late-stage disease or simply markers of evolutionary fitness over time.

Construction and Functional Evaluation of a Three-Dimensional Blood-Brain Barrier Model Equipped With Human Induced Pluripotent Stem Cell-Derived Brain Microvascular Endothelial Cells

Summary: This study constructed and validated an in vivo three-dimensional blood-brain barrier (3D-BBB) model system equipped with brain microvascular endothelial cells derived from human induced pluripotent stem cells (hiPS-BMECs). The 3D capillary structure and expression of tight-junction proteins and transporters were confirmed, and the mRNA expression of transporters in the 3D environment was determined. The 3D-BBB system showed several important characteristics of the human BBB and is expected to be a useful tool in the development of central nervous system drugs.
