Silencing SIRT5 induces the senescence of UCB-MSCs exposed to TNF-α by reduction of fatty acid β-oxidation and anti-oxidation

Summary: In this study, the regulatory effect of tumor necrosis factor-alpha (TNF-α) on the redox regulation of mesenchymal stem cells (MSCs) was investigated. It was found that TNF-α plays a role in protecting MSCs from oxidative stress by increasing fatty acid synthesis and lipid droplet formation. TNF-α also regulates the beta-oxidation of unsaturated fatty acids in lipid droplets, reducing mitochondrial ROS accumulation. Additionally, TNF-α activates SIRT5/Nrf2 signaling to upregulate superoxide dismutase 2 (SOD2), maintaining mitochondrial ROS homeostasis. The findings suggest that SIRT5 plays a central role in protecting TNF-α-induced MSC senescence through FA beta-oxidation and SOD2-mediated antioxidation.
