Genomic Analysis of Waterpipe Smoke-Induced Lung Tumor Autophagy and Plasticity

Rania Faouzi Zaarour et al.

Summary: This study revealed the activation of autophagy in lung cancer cells exposed to waterpipe smoke and investigated its relationship with smoking status in lung adenocarcinoma patients. Smokers exhibited a higher tumor mutational burden, higher CD8+ T-cell level, and lower dysfunction level compared to non-smokers. While there were no relative differences in the expression of immune checkpoint genes between smokers and non-smokers, other genes including B7-1, B7-2, IDO1, and CD200R1 showed higher expression in non-smokers. Therefore, targeting autophagy in smoker patients with lung adenocarcinoma, in combination with checkpoint inhibitor-targeted therapies or chemotherapy, should be considered.

Tumor Hypoxia Regulates Immune Escape/Invasion: Influence on Angiogenesis and Potential Impact of Hypoxic Biomarkers on Cancer Therapies

Raefa Abou Khouzam et al.

Summary: The environmental and metabolic pressures in the tumor microenvironment have a significant impact on tumor development, especially hypoxia-triggered events that promote growth, enhance resistance to the immune response, and stimulate angiogenesis. Normalizing tumor vasculature can restore integrity, increase perfusion, alleviate hypoxia, and reshape anti-tumor immunity, offering potential in achieving stable vascular normalization for mature and functional blood vessels.

Cyclic Hypoxia: An Update on Its Characteristics, Methods to Measure It and Biological Implications in Cancer

Samuel B. Bader et al.

Summary: Hypoxia refers to insufficient levels of oxygen in biological situations, which can occur in various diseases including cancer. Tumor hypoxia is known to lead to more aggressive and difficult-to-treat diseases. Tumor hypoxia is not static, and dynamic changes in oxygen levels have significant biological consequences.

Metabolic support of tumour-infiltrating regulatory T cells by lactic acid

McLane J. Watson et al.

Summary: T-reg cells exhibit broad heterogeneity in glucose metabolism within normal and transformed tissues, utilizing an alternative metabolic pathway to maintain their suppressive function and proliferation. Glucose uptake is correlated with poorer suppressive function and long-term instability, while high-glucose conditions impair T-reg cell function and stability. Additionally, T-reg cells upregulate pathways involved in the metabolism of lactic acid, with lactate uptake being dispensable for peripheral T-reg cells but required intratumorally for slowed tumor growth and increased response to immunotherapy.

Downregulation of both mismatch repair and non-homologous end-joining pathways in hypoxic brain tumour cell lines

Sophie Cowman et al.

Summary: This study investigated the impact of chronic hypoxia on DNA repair gene expression in glioblastoma cell lines, revealing that hypoxia primarily resulted in downregulation of DNA repair genes, with the extent of downregulation depending on the severity of hypoxia. Some downregulations were directly controlled by HIF activity, indicating that multiple molecular mechanisms are involved in hypoxia-induced reprogramming of DNA repair gene transcription.

An Eight-Gene Hypoxia Signature Predicts Survival in Pancreatic Cancer and Is Associated With an Immunosuppressed Tumor Microenvironment

Raefa Abou Khouzam et al.

Summary: An eight-gene hypoxia signature identified in this study showed significant association with overall survival in pancreatic ductal adenocarcinoma (PDA), indicating its potential prognostic value. Tumors with high hypoxia were found to have less immune cell infiltration and lower expression of cytokines and inflammatory markers, suggesting a potential role of the signature in identifying an immune cold, hypoxic tumor microenvironment. The combination of the hypoxia signature with an immune metric revealed worse survival outcomes in patients with high hypoxia and low immune reactivity, highlighting the possible utility of this approach in refining survival estimates.
