Targeting TBK1 to overcome resistance to cancer immunotherapy

Summary: In this study, TBK1 was identified as a candidate immune-evasion gene through a genetic screen. Inhibition of TBK1 enhances the response to PD-1 blockade by reducing the cytotoxicity threshold to cytokines. Targeting TBK1 is an effective strategy to overcome resistance to cancer immunotherapy.

BRAFV600E;K601Q metastatic melanoma patient-derived organoids and docking analysis to predict the response to targeted therapy

Summary: The V600E mutation in BRAF is associated with increased phosphorylation of Erk1/2 and high sensitivity to BRAFi/MEKi combination in metastatic melanoma. However, a tandem mutation in BRAF, V600 and K601, leads to a different response to the combination treatment. Generating BRAF(V600E;K601Q) patient-derived organoids (PDOs) and conducting docking analysis revealed that the tandem mutation reduced the affinity for Vemurafenib. Tumor analysis also showed increased phosphorylation of Erk1/2 and activation of Notch resistance signaling in BRAF(V600E;K601Q).

Preclinical platforms to study therapeutic efficacy of human γδ T cells

Summary: This study reveals the exhausted immunophenotypes and limited anti-tumor capacity of tumor-infiltrating gamma delta T cells in melanoma 3D models. Engineering the Extracellular Matrix for Organoid Culture

Summary: Organoids have great potential in clinical translational research as they provide a close-to-physiological model of native tissues. However, the use of poorly defined matrices has limited their clinical application. Recent studies have emphasized the importance of extracellular matrix composition, cell-matrix interactions, and mechanical signals in the growth and differentiation of organoids. Hydrogel systems, made from natural or synthetic materials, have been designed to recreate the stem cell niche in vitro, providing biochemical, biophysical, and mechanical signals. This review discusses how recapturing various aspects of the tissue-specific environment through matrix design can accelerate the translation of organoid technology from the lab to therapeutic and pharmaceutical applications.

BRAF(V600E;K601Q) metastatic melanoma patient-derived organoids and docking analysis to predict the response to targeted therapy

Summary: The study found that patients with the BRAF(V600E;K601Q) mutation, although sensitive to combination therapy with Vemurafenib and Cobimetinib, were not as responsive as those with a single BRAF(V600E) mutation.
