Mitophagy bridges DNA sensing with metabolic adaption to expand lung cancer stem-like cells

Zhen Liu et al.

Summary: This study identifies hypermitophagy as a characteristic of human lung cancer stem cells (CSCs), promoting metabolic adaption through the Notch1-AMPK axis to drive CSC expansion.

Mitochondrial Fusion: The Machineries In and Out

Song Gao et al.

Summary: Mitochondria are dynamic organelles that undergo fusion and fission, which can impact their function and lead to diseases. Fusion of outer and inner mitochondrial membranes is mediated by dynamin-like proteins, but the molecular mechanisms behind this process remain unclear despite recent structural studies.

Involvement of CDK11B-mediated SPDEF ubiquitination and SPDEF-mediated microRNA-448 activation in the oncogenicity and self-renewal of hepatocellular carcinoma stem cells

Jun-Cheng Guo et al.

Summary: Increasing evidence suggests that cyclin-dependent kinases play a crucial role in hepatocellular carcinoma biology. This study found that CDK11B-mediated ubiquitination influences HCC stem cells. CDK11B degrades SPDEF, which then inhibits DOT1L through miR-448, promoting self-renewal of HCC stem cells.

Mitochondrial oxidative metabolism contributes to a cancer stem cell phenotype in cholangiocarcinoma

Chiara Raggi et al.

Summary: The study reveals that cancer stem cells in cholangiocarcinoma have a more efficient respiratory phenotype, relying on mitochondrial oxidative metabolism and PGC-1a to maintain CSC features. Targeting mitochondrial complex I or PGC-1a in CCA-SPH impairs spherogenicity and expression of markers related to CSC phenotype, pluripotency, and epithelial-mesenchymal transition. Administration of metformin or SR-18292 in mice with CCA-SPH xenografts significantly reduces tumor growth and reverts the phenotype to that of cells grown in monolayer.
