Persistence of improved glucose homeostasis in Gclm null mice with age and cadmium treatment

Christopher M. Schaupp et al.

Summary: Antioxidant signaling is a crucial cellular defense pathway, and its importance in aging has been well-documented. This study investigated the effect of aging on the favorable phenotypes observed in young Gclm null mice, including increased antioxidant activity and improved glucose homeostasis. The results showed that these phenotypes persisted into old age and were associated with transcriptional activation of the Nrf2, AMPK, and PPAR gamma pathways.

PRMT4 promotes ferroptosis to aggravate doxorubicin-induced cardiomyopathy via inhibition of the Nrf2/GPX4 pathway

Yilong Wang et al.

Summary: PRMT4 promotes ferroptosis in DOX-induced cardiomyocyte ferroptosis by interacting with Nrf2 and inhibiting its nuclear translocation, which subsequently suppresses the transcription of GPX4. Activation of Nrf2 or administration of Fer-1 abolishes the detrimental role of PRMT4 in DOX-induced cardiomyocyte ferroptosis.

Targeting ferroptosis in acute kidney injury

Lihua Ni et al.

Summary: Acute kidney injury (AKI) is a major public health problem, and ferroptosis has been found to play an important role in it. The network mechanism of ferroptosis in AKI is complex and multifactorial, and strategies targeting ferroptosis show great potential.

Roles of the Neuron-Restrictive Silencer Factor in the Pathophysiological Process of the Central Nervous System

Xin-Jin Su et al.

Summary: The neuron-restrictive silencer factor (NRSF) is a zinc finger transcription factor that plays a crucial role in the development and function of the nervous system. NRSF can both silence and activate gene transcription, and its dysregulation has been implicated in various brain disorders and tumorigenesis. Targeting NRSF and its co-repressors may hold promise as a therapeutic strategy for neural disorders.

Cancer cells dying from ferroptosis impede dendritic cell-mediated anti-tumor immunity

Bartosz Wiernicki et al.

Summary: Ferroptosis, a form of cell death characterized by iron accumulation and lipid peroxidation, has lower immunogenicity compared to other cell death modalities, inhibiting the maturation and antigen cross-presentation of immune cells and hence weakening the anti-tumor immune response.

Quercetin alleviates acute kidney injury by inhibiting ferroptosis

Yue Wang et al.

Summary: The study demonstrated that quercetin (QCT), a natural flavonoid, inhibited ferroptosis in renal proximal tubular epithelial cells by reducing lipid ROS levels and increasing glutathione (GSH) levels. QCT also ameliorated acute kidney injury induced by ischemia-reperfusion (I/R) or folic acid (FA). Furthermore, RNA-sequence analysis revealed ATF3 as a key factor in QCT's inhibition of ferroptosis. Additionally, QCT inhibited the recruitment of pro-inflammatory macrophages in AKI.

Kidney physiology and susceptibility to acute kidney injury: implications for renoprotection

Holger Scholz et al.

Summary: The authors discuss the specific kidney regions that are at particular risk of injury, and how this information can help in finding new ways to mitigate injury and avoid permanent damage. They suggest that the physiological heterogeneity of the kidney should be considered when exploring novel renoprotective strategies, such as improving kidney tissue oxygenation and modulating cellular energy metabolism.

REST and Stress Resistance in the Aging Kidney

Sato Magassa et al.

Summary: The study identified REST as a central regulator of podocyte adaptation to injury and aging, showing its effects on podocyte cytoskeleton to promote resistance to mechanical stressors and augment podocyte survival. The upregulation of REST expression in aging human podocytes suggests a conserved mechanism of stress resistance and potential therapeutic implications.

Ferroptosis: mechanisms and links with diseases

Hong-fa Yan et al.

Summary: Ferroptosis is a form of cell death that is iron-dependent and involves accumulation of lethal lipid species from lipid peroxidation. This review discusses the regulatory mechanisms of ferroptosis and its connections with iron, lipid, and cysteine metabolism pathways. Additionally, it explores the role of ferroptosis in diseases such as cancer, ischemia/reperfusion, Alzheimer's disease, and Parkinson's disease, and evaluates the potential therapeutic applications of ferroptosis inhibitors in clinical settings.
