Long non-coding RNAs: definitions, functions, challenges and recommendations

John S. S. Mattick et al.

Summary: Genes encoding lncRNAs are abundant in complex organisms and are transcribed by RNA polymerase I, II, and III, as well as processed introns. The classification and annotation of lncRNAs are challenging due to their various functions, isoforms, and interactions with other genes. These lncRNAs evolve rapidly, are cell type-specific, and regulate multiple cellular processes, including gene expression and translation control.

GATA6-AS1 Regulates Intestinal Epithelial Mitochondrial Functions, and its Reduced Expression is Linked to Intestinal Inflammation and Less Favourable Disease Course in Ulcerative Colitis

Katya E. Sosnovski et al.

Summary: The study found that the levels of GATA6-AS1, a long non-coding RNA, were reduced in inflammatory bowel disease. The reduction of GATA6-AS1 was associated with impaired epithelial mitochondrial functions. The study also identified the interactome of GATA6-AS1, which was enriched for mitochondrial proteins.

Clinical and Host Biological Factors Predict Colectomy Risk in Children Newly Diagnosed With Ulcerative Colitis

Jeffrey S. Hyams et al.

Summary: This study developed a model for predicting the risk of colectomy in children newly diagnosed with ulcerative colitis. The findings suggest that a small group of children with severe UC still require colectomy despite current therapies. The gene signature observations provide additional targets for managing patients who do not respond to current medical treatments.

Identification of gut microbial species linked with disease variability in a widely used mouse model of colitis

Samuel C. Forster et al.

Summary: A study on a mouse model of inflammatory bowel disease reveals that the gut microbiome plays a crucial role in the variability across genetically identical mice, and identifies two bacterial species that affect treatment outcomes differently. This finding is important for the improvement of mouse models for human intestinal diseases.

The HASTER lncRNA promoter is a cis-acting transcriptional stabilizer of HNF1A

Anthony Beucher et al.

Summary: This study reveals that HASTER plays a role in maintaining the normal concentration of HNF1A through positive and negative feedback loops. Mutations in Haster lead to silencing or overexpression of HNF1A in pancreatic beta cells, resulting in hyperglycemia. The HASTER-dependent negative feedback is essential for preventing inappropriate binding of HNF1A to genomic regions.

Meta-analysis defines predominant shared microbial responses in various diseases and a specific inflammatory bowel disease signal

Haya Abbas-Egbariya et al.

Summary: This study reveals a common non-specific response of the gut microbiome in a wide array of diseases, which may cause confusion in disease classification. A universal dysbiosis index is developed to prioritize fecal samples and donors across different diseases. Additionally, specific bacteria associated with inflammatory bowel disease are identified as potential biomarkers and intervention targets.

Gene regulation by long non-coding RNAs and its biological functions

Luisa Statello et al.

Summary: Recent studies have shed new light on the biogenesis and functions of long non-coding RNAs (lncRNAs), showcasing their diverse roles in gene regulation and signaling pathways, particularly in the contexts of neuronal disorders, immune responses, and cancer. The discovery of unique biogenesis pathways and subcellular localizations of lncRNAs has opened up potential therapeutic avenues for targeting lncRNAs in various biological and pathophysiological conditions.

Mucosal Genomics Implicate Lymphocyte Activation and Lipid Metabolism in Refractory Environmental Enteric Dysfunction

Yael Haberman et al.

Summary: The study found that Environmental Enteric Dysfunction (EED) leads to the suppression of antioxidant, detoxification, and lipid metabolism genes, while inducing the expression of antimicrobial response, interferon, and lymphocyte activation genes. EED also shows hypermethylation of epithelial metabolism and barrier function genes, and hypomethylation of immune response and cell proliferation genes. In addition, EED is associated with lymphocyte proliferation, expression of epithelial metabolism genes, and severity of histological damage, fecal energy loss, and malnutrition.

Testicular germ cell-specific lncRNA, Teshl, is required for complete expression of Y chromosome genes and a normal offspring sex ratio

Seong Hyeon Hong et al.

Summary: The study reveals that the testicular germ cell-specific lncRNA NR_038002 interacts with HSF2 to regulate the expression of MSYq genes, affecting male mouse fertility and offspring sex ratio. This discovery provides new insights into the molecular mechanism of testis-specific activity.
