Pyrazolone derivatives as potent and selective small-molecule SIRT5 inhibitors

Summary: In this study, a novel SIRT5-selective inhibitor was identified and optimized, resulting in compound 47 with 100-fold improved potency. Summary: A metal- and photosensitizer-free protocol for decarboxylative C-H (amino)alkylation of heteroarenes using carboxylic acids as reagents has been developed. The reaction is performed in a sustainable solvent (dimethyl carbonate, DMC) and is tolerant to various carboxylic acids, providing the target products in acceptable yields. Importantly, this approach can efficiently convert amino acids and carboxyl-containing drugs into desired products, offering a green and efficient method for the conversion of abundant feedstock materials into medically relevant drug-like moieties.

Visible-light-induced direct perfluoroalkylation/heteroarylation of [1.1.1]propellane to diverse bicyclo[1.1.1]pentanes (BCPs) under metal and photocatalyst-free conditions

Summary: A green and novel multi-component reaction for direct perfluoroalkylation/heteroarylation of [1.1.1]propellane with heteroarenes and perfluoroalkyl iodines is described. This transformation is facilitated by visible light in the absence of any transition-metal catalysts and photocatalysts. Various heteroarenes and perfluoroalkyl iodines are well compatible, providing the corresponding products in moderate-to-good yields. The control experiments demonstrate that a radical-relay mechanism is responsible for the cascade reaction. Such a methodology offers an efficient and practical access to diverse 1-perfluoroalkyl-3-heteroaryl bicyclo[1.1.1]pentanes (BCPs) with the potential for various applications in organic synthesis.

TBAI/H2O-cooperative electrocatalytic decarboxylation coupling-annulation of quinoxalin-2(1 H )-ones with N-arylglycines

Summary: The first example of TBAI/H2O cooperative electrocatalytic coupling-annulation of quinoxalin-2(1H)-ones with N-arylglycines was developed. A broad range of tetrahydroimidazo[1,5-a]quinoxalin-4(5H)-ones were obtained in good to excellent yields with exclusive chemoselectivities and excellent regioselectivities. The H-hydrogen bond served as a key factor for the electrocatalytic production of aminomethyl radical at lower oxidative potential.

Selective Mono- and Diamination of Ketones in a Combined Copper-Organocatalyst System

Electrochemical Decarbonylative Aminosulfonylation of Alkynes with Sulfinates and N-(Formyl)anilides

Summary: An unprecedented electrochemical three-component reaction involving phenylacetylene, sulfinate, and N-(formyl)anilide has been discovered. The reaction can be carried out in an undivided cell with a graphite anode and cathode using tetrabutylammonium iodide as an electrolyte, and the addition of silver(I) oxide and catalytic amounts of iodine significantly promotes the reaction. The transformation can also be conducted under photoredox-catalyzed conditions.

Summary: In this study, an metal-free, exogenous-oxidant-free, and mild conditions electrochemical synthesis of polysubstituted sulfonated pyrazoles was developed. NH2-functionalized enaminones or N,N-disubstituted enaminones reacted with aryl/alkyl sulfonyl hydrazides to produce tetra- or trisubstituted sulfonated pyrazoles in moderate to good yields, respectively. The gram-scale electro-chemical transformation demonstrated the efficiency and practicability of this method. The formation mechanism involves intermolecular condensation, radical-radical cross coupling sulfonylation, and pyrazole annulation.

Electrooxidative tandem cyclization of enaminones to give 3-arylthiochromone derivatives
