Structural basis of GABARAP-mediated GABAA receptor trafficking and functions on GABAergic synaptic transmission

Jin Ye et al.

Summary: GABARAP and GABARAPL1 stabilize GABA(A) receptors by directly binding to the γ 2 subunit, promoting receptor trafficking pathways, and modulating receptor localization at synapses.

ATG4D is the main ATG8 delipidating enzyme in mammalian cells and protects against cerebellar neurodegeneration

Isaac Tamargo-Gomez et al.

Summary: Despite significant advances in autophagy research in recent years, the specific functions of the four mammalian Atg4 proteases (ATG4A-D) remain unclear. The study shows that ATG4D is the main delipidating enzyme and its loss leads to accumulation of membrane-bound forms of mATG8s, increased cellular autophagosome number, and reduced autophagosome average size in mammalian cells. Loss of ATG4D in mice results in cerebellar neurodegeneration and impaired motor coordination due to alterations in trafficking/clustering of GABA(A) receptors, emphasizing the neuroprotective role of ATG4D in mammals.

Non-canonical autophagy drives alternative ATG8 conjugation to phosphatidylserine

Joanne Durgan et al.

Summary: Autophagy is a crucial cellular process mediated by ATG8 protein lipidation to PE or PS, which are differentially delipidated by the ATG4 family, showing distinct cellular dynamics; this reveals important molecular differences in the autophagy pathway.

ATG4 family proteins drive phagophore growth independently of the LC3/GABARAP lipidation system

Thanh Ngoc Nguyen et al.

Summary: The ATG4 family of proteins promote autophagosome formation independently of ATG8 family processing and protease activity, and also have a role in transporting ATG9A vesicles to mitochondria with proximity partners like LRBA. Additionally, ATG4s facilitate phagophore-ER contacts during lipid-transfer phase of autophagosome formation and act as deubiquitinating-like enzymes in disassembling ATG8-protein conjugates.
