Arabic gum plus colistin coated moxifloxacin-loaded nanoparticles for the treatment of bone infection caused by Escherichia coli

J. J. Aguilera-Correa et al.

Summary: This study developed a nanomedicine using mesoporous silica nanoparticles to target and eradicate E. coli-related bone infections. In vitro and in vivo experiments showed that the nanosystem effectively targeted E. coli biofilm and displayed strong antibacterial properties, offering a promising treatment option for bone infections caused by enterobacteria like E. coli.

Why 90 % of clinical drug development fails and how to improve it?

Duxin Sun et al.

Summary: Despite the implementation of many successful strategies, the majority of clinical drug development fails. This study suggests that current drug optimization strategies overlook tissue exposure/selectivity, leading to misleading drug candidate selection and impacting clinical efficacy/toxicity balance. The proposed structure-tissue exposure/selectivity-activity relationship (STAR) classification system aims to improve drug optimization and enhance the success rate of clinical drug development.

Synthesis of a theranostic platform based on fibrous silica nanoparticles for the enhanced treatment of triple-negative breast cancer promoted by a combination of chemotherapeutic agents

Karina Ovejero-Paredes et al.

Summary: A new series of theranostic silica materials based on fibrous silica particles have been successfully prepared and characterized. These materials show synergistic therapeutic activity and targeting ability against triple negative breast cancer. In vivo and in vitro studies confirm the promising applicability of these silica-based nanoplatforms for advanced drug delivery and diagnosis.

Folate Functionalized Lipid Nanoparticles for Targeted Therapy of Methicillin-Resistant Staphylococcus aureus

Kushal Vanamala et al.

Summary: This study aims to improve the efficacy of VAN treatment by identifying the folate receptor for MRSA infected tissues and developing folate decorated lipid nanoparticles containing VAN (LVAN). In comparison to conventional VAN, LVAN showed a higher bactericidal effect and a superior ability to inhibit biofilm in MRSA with an enhanced accumulation in MRSA infected thigh tissues and a reduced accumulation in kidney. The results suggested that LVAN is a promising candidate to overcome the current limitations of bacterial resistance and adverse side effects in kidneys found in VAN.

Evaluation of metal-based antimicrobial compounds for the treatment of bacterial pathogens

Andris Evans et al.

Summary: Antimicrobial resistance (AMR) is a major global health challenge, with rising prevalence worldwide. The lack of antibiotics with novel modes of action hinders effective treatment of bacterial infections with resistance. Metal-based antibacterial compounds offer a promising solution for addressing this issue.

Burns and biofilms: priority pathogens and in vivo models

Evgenia Maslova et al.

Summary: Burn wounds can severely damage human skin and lead to infections, which are the leading cause of death among burn wound patients. Surviving patients may face difficulties in treating infections, causing delayed healing and prolonged hospital stays. Biofilm formation by bacteria in burn wounds contributes to treatment failure and mortality, as it makes the bacteria more resistant to disinfection and immune responses.
