Evaluation of the Cytotoxic Effect of Pd(2)Spm against Prostate Cancer through Vibrational Microspectroscopies

Raquel C. C. Laginha et al.

Summary: In this study, the cytotoxic effects of a complex with two Pd(II) centers (Pd(2)Spm) on healthy and cancerous prostate human cell lines were evaluated and compared to cisplatin. Fourier Transform Infrared (FTIR) and Raman microspectroscopies were used to understand the molecular mechanisms of action of both drugs, revealing differences in their impact on DNA, lipids, and proteins.

Metal complexes induced ferroptosis for anticancer therapy

Shumeng Li et al.

Summary: Metal complexes for anti-tumor treatment have developed rapidly, while some tumor cells are resistant to apoptosis and not sensitive to metallodrugs that function through the apoptotic pathway. Recently, metal complexes have been reported to induce ferroptosis against tumor cells, providing new opportunities for anticancer therapy. This perspective introduces ferroptosis-inducing metal complexes, their working mechanisms, and discusses the challenges and opportunities.

Pd2Spermine Complex Shows Cancer Selectivity and Efficacy to Inhibit Growth of Triple-Negative Breast Tumors in Mice

Martin Vojtek et al.

Summary: This study evaluated the anticancer effects of Pd(2)Spm on triple-negative breast cancer and compared it with cisplatin. Results showed that Pd(2)Spm exhibited promising selective antiproliferative activity, inhibited migration and angiogenesis, and demonstrated significant antitumor effects in vivo, with lower systemic toxicity compared to cisplatin.

Impact of the Pd(2)Spm (Spermine) Complex on the Metabolism of Triple-Negative Breast Cancer Tumors of a Xenograft Mouse Model

Tatiana J. Carneiro et al.

Summary: Interest in palladium(II) compounds as potential new anticancer drugs has increased due to their high toxicity and acquired resistance to platinum(II)-derived agents, such as cisplatin. This study examines the in vivo response of TNBC tumors to a Pd(2)Spm complex compared to cisplatin, showing distinct impacts on polar metabolites but not on tumor lipophilic metabolism. The results suggest that the Pd(2)Spm complex triggers more variations in certain metabolites related to cytotoxicity and may have a distinct mechanism of action in TNBC metabolism.

Preclinical Pharmacokinetics and Biodistribution of Anticancer Dinuclear Palladium(II)-Spermine Complex (Pd(2)Spm) in Mice

Martin Vojtek et al.

Summary: This study assessed the pharmacokinetics and biodistribution of a dinuclear palladium(II)-spermine chelate (Pd(2)Spm) in mice, showing promising characteristics for its potential use as an anticancer agent. Palladium exhibited biphasic kinetics in serum, with high tissue distribution including in the kidney, liver, lungs, ovaries, adipose tissue, and mammary glands. These findings suggest Pd(2)Spm may be a promising pharmacological agent for cancer treatment.

Survival benefit of platinum-based regimen in early stage triple negative breast cancer: A meta-analysis of randomized controlled trials

Lei Bian et al.

Summary: Platinum-based chemotherapy regimens have been shown to significantly improve disease-free survival (DFS) and overall survival (OS) in early stage triple negative breast cancer (eTNBC) treatment. Whether adding platinum to standard anthracyclines & taxanes based regimens or combining platinum with taxanes alone, both strategies demonstrate consistent survival benefits in eTNBC patients. The present meta-analysis recommends platinum-based regimens, particularly the A&T + Pt mode, for eTNBC patients, if tolerable toxicities, to potentially revolutionize TNBC therapy.
