High-Performance Delivery of a CRISPR Interference System via Lipid-Polymer Hybrid Nanoparticles Combined with Ultrasound-Mediated Microbubble Destruction for Tumor-Specific Gene Repression

Summary: In this study, a combination of pH-responsive lipid-polymer hybrid nanoparticles (PLPNs) and ultrasound-mediated microbubble destruction (UMMD) is used for the efficient delivery of the CRISPRi system. The PLPNs loaded with the CRISPRi plasmid can effectively target cancer cells and increase the time spent in circulation in vivo. Furthermore, the combination of PLPNs with UMMD achieves a higher cellular uptake and retention of the CRISPRi plasmid. When applied to breast cancer, this system effectively inhibits miR-10b and disturbs multiple cell migration and invasion-related signaling pathways, resulting in a significant inhibition of lung metastasis.

Towards controlled drug delivery in brain tumors with microbubble-enhanced focused ultrasound

Early clinical studies have demonstrated safety, and ongoing Phase I/II trials are testing the preclinical findings.

Summary: This research focuses on overcoming biological barriers for therapeutic nanocarriers through the use of switchable nanocarriers that are responsive to both endogenous and exogenous stimuli. By combining endogenous responsiveness with exogenous ultrasound responsiveness, the nanocarriers are able to balance the conflicting requirements for prolonged blood circulation and deep tumor penetration. The results show that the ultrasound-responsive stiffness of the nanogel allows for enhanced deep tumor therapy.

Dual-responsive nanodroplets combined with ultrasound-targeted microbubble destruction suppress tumor growth and metastasis via autophagy blockade

Summary: The inhibition of autophagy is a feasible clinical strategy in tumor therapy. In this study, hydroxychloroquine-loaded nanodroplets (HCQ-NDs) were developed to overcome the limitations of traditional autophagy inhibitors. The combination of ultrasound targeted microbubble destruction (UTMD) and HCQ-NDs effectively disrupted the tumor cell homeostasis, blocking autophagic flux and showing strong synergistic antitumor efficacy. HCQ-NDs also exhibited excellent tumor-targeting, biocompatibility, biosafety, and contrast-enhanced ultrasound imaging properties.
