Nanoemulgel: A Novel Nano Carrier as a Tool for Topical Drug Delivery

Mahipal Reddy Donthi et al.

Summary: Nano-emulgel is a novel drug delivery system that aims to enhance the therapeutic effectiveness of lipophilic drugs. It addresses the limitations of lipophilic formulations such as poor solubility, unpredictable absorption, and low oral bioavailability. By incorporating nano-emulsion into gel, the stability and drug delivery for immediate and controlled release are improved. The focus on nano-emulgel has increased due to its targeted delivery, ease of application, absence of gastrointestinal degradation or the first pass metabolism, and safety profile. This review discusses the formulation components, pharmacokinetics, and safety profiles of nano-emulgel for topical drug delivery.

Revisiting techniques to evaluate drug permeation through skin

Vamshi Krishna Rapalli et al.

Summary: Investigating methods for drug penetration through skin layers include various techniques such as in vitro diffusion, ex vivo isolated perfused skin models, and in vivo tape stripping, which need to be integrated at various stages of formulation development to consider various effects. The existing methodological gap requires further research to improve the reliability of skin permeation experimental data.

Combination of subtherapeutic anti-TNF dose with dasatinib restores clinical and molecular arthritogenic profiles better than standard anti-TNF treatment

Lydia Ntari et al.

Summary: The study showed that the Tyrosine Kinase inhibitors dasatinib and bosutinib have therapeutic effects on TNF-dependent arthritis in mice. Dasatinib, either as a monotherapy or in combination with a subtherapeutic dose of anti-hTNF biologics, exhibited significant improvement in clinical and histopathological signs of the disease. Combination therapy with dasatinib and anti-hTNF agents proved to have a distinct mode of action in restoring the arthritogenic gene signature, potentially offering an alternative to high-dose anti-hTNF monotherapy and increasing the number of patients responding to treatment.
