CT texture analysis in predicting treatment response and survival in patients with hepatocellular carcinoma treated with transarterial chemoembolization using random forest models

He An et al.

Summary: This study used texture features derived from contrast-enhanced CT, general imaging features, and clinical information to predict treatment response and survival in HCC patients treated with TACE. The results showed that the random forest algorithm based on texture features, combined with general imaging features and clinical information, can effectively predict prognosis in HCC patients.

T-cell exhaustion signatures characterize the immune landscape and predict HCC prognosis via integrating single-cell RNA-seq and bulk RNA-sequencing

Hao Chi et al.

Summary: A TEX-based signature was developed using single-cell RNA-seq and high-throughput RNA sequencing to evaluate the prognosis and immunotherapeutic response of HCC patients. The study found that TEX genes were closely associated with HCC prognosis, and patients in the low-risk group had a better overall survival rate. This study provides a new method for assessing immune therapy efficacy and has important implications for future precision immuno-oncology studies.

Treatment and the prognosis of hepatocellular carcinoma in Asia

Takuji Torimura et al.

Summary: Hepatocellular carcinoma is the most common malignant tumor in Asia, and treatment methods are determined based on tumor staging and liver function. Treatment guidelines in Asian countries are generally more aggressive and include combination therapies and alternative treatments. Molecular targeted agents, antibodies, and immune checkpoint inhibitors have been used for the treatment of hepatocellular carcinoma. Taiwan and Japan have the best clinical outcomes for patients with hepatocellular carcinoma.

Preoperative Prognostic Nutritional Index and Neutrophil-to-Lymphocyte Ratio Predict Survival Outcomes of Patients With Hepatocellular Carcinoma After Curative Resection

Zhen Qu et al.

Summary: This study analyzed the data of 215 patients who underwent curative resection for hepatocellular carcinoma (HCC) and found that prognostic nutritional index (PNI) and neutrophil-to-lymphocyte ratio (NLR) were independent prognostic factors for overall survival and recurrence-free survival. A new nomogram based on PNI and NLR may provide individualized predictions of survival and recurrence for HCC patients.

Artificial intelligence for the prevention and clinical management of hepatocellular carcinoma

Julien Calderaro et al.

Summary: Hepatocellular carcinoma (HCC) is a growing malignancy with increasing incidence and mortality rates globally. Artificial intelligence (AI) provides a unique opportunity to enhance HCC risk prediction, diagnosis, and prognostication. Machine learning (ML) and deep learning (DL) models, applied to diverse data sources, have shown promising results in improving accuracy of HCC risk prediction, detection, and treatment response prediction. However, further research is needed to address challenges related to standardizing AI data and ensuring interpretability of results.

Ensemble learning based on efficient features combination can predict the outcome of recurrence-free survival in patients with hepatocellular carcinoma within three years after surgery

Liyang Wang et al.

Summary: This study proposes a machine learning-based method to predict the recurrence outcome of hepatocellular carcinoma patients within three years after surgery by analyzing preoperative imaging and clinical data. The results show that the method can relatively accurately predict recurrence and assist physicians in evaluating patients before surgery.

The Preoperative Prognostic Nutritional Index in Hepatocellular Carcinoma After Curative Hepatectomy: A Retrospective Cohort Study and Meta-Analysis

Xiaoxiao Fan et al.

Summary: The study demonstrated that a low preoperative PNI was associated with poor recurrence-free survival in HCC patients after hepatectomy. Furthermore, a low PNI was found to be an independent prognostic factor for overall survival and recurrence-free survival rates in HCC patients, especially those with TNM stage I. The pooled analysis also indicated a significant association between low PNI and poor overall survival and recurrence-free survival rates.

Exploring prognostic indicators in the pathological images of hepatocellular carcinoma based on deep learning

Jie-Yi Shi et al.

Summary: The study proposed a deep learning framework for analysing hepatocellular carcinoma (HCC) pathological images and exploring new prognostic phenotypes. Through TRS scoring and risk activation mapping, it effectively evaluated patient outcomes and identified the main underlying features of TRS. The results showed that TRS is an independent prognosticator superior to clinical staging systems.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Jiarui Yang et al.

Summary: pa-TACE, postoperative radiotherapy, and sorafenib can improve the prognosis of patients with hepatocellular carcinoma with microvascular invasion after curative resection compared to postoperative conservative treatment. Postoperative radiotherapy can reduce the recurrence of patients with HCC with MVI after curative resection when compared to pa-TACE.

Development and Validation of a Machine Learning Prognostic Model for Hepatocellular Carcinoma Recurrence After Surgical Resection

Yao Huang et al.

Summary: A machine learning prognostic model was developed to identify high-risk patients after surgical resection, with variable importance reviewed in different time intervals. Tumor characteristics contribute more to HCC relapse in 0 to 1 year, while HBV infection and smoking largely affect patients' outcome in 3 to 5 years.

Summary: The international cohort study found that the ERASL models had lower discriminatory power for Western patients compared to Japanese patients, and recalibration was needed to improve prediction accuracy. A model specifically tailored for Western patients or explaining the East-West difference is still necessary to be developed.
