Secreted mutant calreticulins as rogue cytokines in myeloproliferative neoplasms

Christian Pecquet et al.

Summary: Mutant CALR proteins bind to and activate the TpoR in cells, driving the development of myeloproliferative neoplasms. These mutant CALR proteins can be found in patient plasma complexed with sTFR1, which increases their stability. They can specifically interact with TpoR on target cells and promote thrombopoietin-independent colony formation.

Life histories of myeloproliferative neoplasms inferred from phylogenies

Nicholas Williams et al.

Summary: By sequencing the clones of 12 patients with myeloproliferative neoplasms, we identified that early driver mutations are the foundation for tumor development and are linked to lifelong growth and evolution. This discovery provides opportunities for early intervention and a new model for cancer development.

The longitudinal dynamics and natural history of clonal haematopoiesis

Margarete A. Fabre et al.

Summary: Clonal haematopoiesis, a common phenomenon in human tissues during aging, was studied by tracking 697 clones from 385 individuals aged 55 years or older over a median of 13 years. The study found that 92.4% of the clones expanded at a stable exponential rate, with different mutations driving different growth rates. Clones with the same mutation had growth rates differing by approximately +/- 5% per year. The study also revealed that mutations driving faster clonal growth carried a higher risk of malignant progression.

In utero origin of myelofibrosis presenting in adult monozygotic twins

Nikolaos Sousos et al.

Summary: Lineage tracing analysis of twins with myelofibrosis suggests in utero transplacental transmission of the tumorigenic clone. The study also reveals the fitness advantage of MPN-associated driver mutations in hematopoietic stem cells.

Clonal haematopoiesis of indeterminate potential and impaired kidney function-A Danish general population study with 11 years follow-up

Morten K. Larsen et al.

Summary: The relationship between clonal haematopoiesis of indeterminate potential (CHIP) and impaired kidney function is unclear. A study in Denmark found that individuals with CALR mutations, especially CALR type 2, had impaired kidney function compared to those without CHIP.

Inferring the initiation and development of myeloproliferative neoplasms

Gurvan Hermange et al.

Summary: This study investigates the initiation and dynamics of blood cancer through mathematical modeling and statistical inference. The findings suggest that CALR(m) mutations tend to occur later in life compared to JAK2(V617F), confirming the higher proliferative advantage of the CALR(m) malignant clone.

The pathobiology of thrombosis, microvascular disease, and hemorrhage in the myeloproliferative neoplasms

Hans Carl Hasselbalch et al.

Summary: Thrombotic, vascular, and bleeding complications are common causes of morbidity and mortality in Philadelphia chromosome-negative myeloproliferative neoplasms (MPNs). Abnormalities in circulating cells in MPNs create a prothrombotic environment, leading to venous, arterial, and microvascular thrombosis. The inflammatory state of MPN stem cells amplifies the clinical thrombotic tendency and promotes further MPN stem cell clonal expansion.

Reconstructing the Lineage Histories and Differentiation Trajectories of Individual Cancer Cells in Myeloproliferative Neoplasms

Debra Van Egeren et al.

Summary: The JAK2-V617F mutation affects the self-renewal and differentiation dynamics of HSCs in MPNs patients, helping to reconstruct cancer lineages, with the mutation likely occurring several decades before diagnosis and providing a selective advantage for mutant HSCs.

Therapeutic Cancer Vaccination With a Peptide Derived From the Calreticulin Exon 9 Mutations Induces Strong Cellular Immune Responses in Patients With CALR-Mutant Chronic Myeloproliferative Neoplasms

Jacob Handlos Grauslund et al.

Summary: In a phase I clinical vaccination trial of CALRLong36 peptide vaccines for CALR-mutant MPN patients, the vaccines were found to be safe and tolerable, and were able to induce T-cell responses. However, the vaccines did not lead to clinical responses, but the study indicates that vaccines targeting mutant CALR may have potential in enhancing anti-tumor immune responses.
