Tumor Mutational Burden Associated With Response to Hyperthermic Intraperitoneal Chemotherapy

Lisi Zeng et al.

Summary: This study found that gastric cancer patients with a higher tumor mutational burden (TMB) have a better response to hyperthermic intraperitoneal chemotherapy (HIPEC) and longer survival time. The results provide important insights for prognostic stratification of HIPEC treatment.

Fibroblast growth factor receptors in cancer: genetic alterations, diagnostics, therapeutic targets and mechanisms of resistance

Summary: FGFRs are aberrantly activated in 5-10% of human cancers through genetic alterations, with FGFR-targeted therapies showing significant advancements in the past decade. However, acquired drug resistance leading to cancer progression remains a challenge in most patients.

Cytoreductive surgery plus hyperthermic intraperitoneal chemotherapy versus cytoreductive surgery alone for colorectal peritoneal metastases (PRODIGE 7): a multicentre, randomised, open-label, phase 3 trial

Francois Quenet et al.

Summary: This study aimed to evaluate the specific benefit of adding hyperthermic intraperitoneal chemotherapy (HIPEC) to cytoreductive surgery compared with receiving cytoreductive surgery alone in patients with colorectal peritoneal metastases. The results showed no significant overall survival benefit after adding HIPEC, with more frequent postoperative late complications, suggesting that cytoreductive surgery alone should be prioritized as a treatment strategy.

The role of molecular biomarkers in outcomes and patient selection for cytoreductive surgery and hyperthermic intraperitoneal chemotherapy for peritoneal metastases of colorectal origin

Daniel Solomon et al.

Summary: This study retrospectively reviewed patients with peritoneal metastases of colorectal cancer who underwent CRS/HIPEC and found that mBRAF mutation was independently associated with earlier recurrence, leading to poor median DFS. Strict patient selection is advisable for these patients.

A Phase Ib/II Study of Pepinemab in Combination with Avelumab in Advanced Non-Small Cell Lung Cancer

Michael R. Shafique et al.

Summary: The combination of pepinemab with avelumab was well tolerated and showed signs of antitumor activity in patients with advanced non-small cell lung cancer, including those who were resistant to immunotherapy and had PD-L1-negative/low tumors. Overall, the combination therapy demonstrated promising safety and efficacy outcomes in this study.

Summary: High-throughput -omics analysis provides a broader understanding of cancer biology, but studies on peritoneal metastases and pseudomyxoma peritonei revealed variability in mutation frequencies and challenges in interpreting results due to small and heterogeneous cohorts.

Summary: In this study, while HIPEC with carboplatin achieved a higher rate of complete tumor resection, it did not show superior clinical outcomes compared to standard treatment, challenging the use of carboplatin HIPEC during secondary cytoreductive surgery for platinum-sensitive recurrent ovarian cancer.
