Ceramide Synthase 1 Inhibits Brain Metastasis of Non-Small Cell Lung Cancer by Interacting with USP14 and Downregulating the PI3K/AKT/mTOR Signaling Pathway

Yiquan Xu et al.

Summary: This study found that brain metastasis (BM) is common and associated with poor prognosis in patients with non-small cell lung cancer (NSCLC). Ceramide synthase 1 (CERS1) plays a role in malignancy development, and its upregulation is associated with better prognosis in NSCLC BM. Furthermore, CERS1 interacts with ubiquitin-specific protease 14 (USP14) and inhibits BM progression by downregulating the PI3K/AKT/mTOR signaling pathway.

CDKN2A/B co-deletion is associated with increased risk of local and distant intracranial recurrence after surgical resection of brain metastases

Ramin A. Morshed et al.

Summary: This study found that CDKN2A/B co-deletion detected in brain metastases is associated with increased central nervous system (CNS) recurrence after surgical resection. Further research is needed to determine whether more aggressive treatment in patients with this mutation may improve outcomes.

Review of Current Principles of the Diagnosis and Management of Brain Metastases

Alex W. Brenner et al.

Summary: Brain metastases, the most common intracranial tumors, can be diagnosed through clinical examination and magnetic resonance imaging. Treatment options include surgery, radiotherapy, and systemic medical therapy, and recent advancements have led to improved performance and survival rates. Whole brain radiotherapy and stereotactic radiosurgery are supported by extensive evidence, while modern immunotherapeutic agents show promise.

The aberrant cancer metabolic gene carbohydrate sulfotransferase 11 promotes non-small cell lung cancer cell metastasis via dysregulation of ceruloplasmin and intracellular iron balance

Wei-Min Chang et al.

Summary: This study identified metabolic genes associated with EMT in lung cancer cells and revealed the role of the CHST11-CP-iron axis in enhancing EMT and promoting metastasis in NSCLC, providing a potential new therapeutic target for NSCLC patients.

Non-Small-Cell Lung Cancer in 2022: A Review for General Practitioners in Oncology

Hamid Mithoowani et al.

Summary: This article presents a comprehensive review of the current best practices in the treatment of non-small cell lung cancer in Canada, covering screening, management of early stage and locally-advanced disease, as well as management of patients with metastatic disease. It also focuses on the incorporation of immunotherapy into practice and its associated toxicities.

Contribution of resident and circulating precursors to tumor-infiltrating CD8(+) T cell populations in lung cancer

Paul Gueguen et al.

Summary: Tumor-infiltrating lymphocytes, especially CD8(+) TILs, serve as a favorable prognostic factor in non-small cell lung cancer. The study used single-cell RNA and TCR sequencing to investigate the functional organization of TIL populations in primary NSCLC, identifying two CD8(+) TIL subpopulations expressing memory-like gene modules and showing their differentiation pathways in the tumor microenvironment.

Multi-site tumor sampling highlights molecular intra-tumor heterogeneity in malignant pleural mesothelioma

Clement Meiller et al.

Summary: This study investigated intra-tumor heterogeneity in malignant pleural mesothelioma (MPM) using the largest series to date and multi-omics profiling of a substantial series of multi-site tumor samples. The results revealed patient-dependent spatial heterogeneity at genetic, transcriptomic, and epigenetic levels, as well as changes in the immune landscape of the tumor microenvironment, supporting the need for multi-sampling for precision medicine.
