SOX4 maintains the stemness of cancer cells via transcriptionally enhancing HDAC1 revealed by comparative proteomics study

Jingshu Liu et al.

Summary: Overexpression of SOX4 promotes stemness of colorectal cancer cells by upregulating CSC markers and inducing proteome changes, including activation of HDAC1 as a key mechanism. The SOX4-HDAC1 axis is conserved in multiple types of cancer, suggesting HDAC1 is a potential drug target for eliminating SOX4-driven human CSCs.

Whole-genome analysis of Nigerian patients with breast cancer reveals ethnic-driven somatic evolution and distinct genomic subtypes

Naser Ansari-Pour et al.

Summary: Research reveals higher genomic instability and intra-tumoral heterogeneity in Nigerian breast cancer patients, as well as early clonal GATA3 mutations and a unique INDEL signature associated with African ancestry. Characterizing tumors for homologous recombination deficiency is shown to have significant clinical relevance, underscoring the importance of including diverse populations in biomedical research. This study sheds light on the unknown breast cancer heterogeneity and tumour evolutionary trajectories among women of African ancestry.

Breast cancer as an example of tumour heterogeneity and tumour cell plasticity during malignant progression

Fabiana Luond et al.

Summary: Tumour heterogeneity, arising from intrinsic and extrinsic factors, has negative impacts on therapy response and prognosis. Cancer cell plasticity enables rapid adaptation to microenvironmental cues, playing a critical role in tumour progression and heterogeneity.

The hippo pathway: A master regulator of liver metabolism, regeneration, and disease

Anh Thu Nguyen-Lefebvre et al.

Summary: The liver is the only visceral organ in the body with a tremendous capacity to regenerate in response to insults. Liver regeneration is a complicated process involving activation of non-parenchymal cells and hepatocytes proliferation. Understanding the mechanisms that control liver regeneration is critical for therapies and patient care.

Subclonal heterogeneity and evolution in breast cancer

Ioanna Mavrommati et al.

Summary: This review focuses on the recent advances in understanding the epigenetic and transcriptomic changes in breast cancer at the single-cell level, and highlights the utility of single-cell tracing and molecular barcoding methodologies in assessing disease evolution and therapy response. Integrating single-cell profiling from patient samples with results from preclinical models can untangle complexities of the disease and identify biomarkers for disease progression, potentially uncovering new targetable vulnerabilities in breast cancer.

ZSCAN16-AS1 expedites hepatocellular carcinoma progression via modulating the miR-181c-5p/SPAG9 axis to activate the JNK pathway

Jianwen Liu et al.

Summary: ZSCAN16-AS1 was significantly up-regulated in HCC cells. Its silencing inhibited HCC cell proliferation, migration, and invasion, while accelerating cell apoptosis. ZSCAN16-AS1 modulated the miR-181 c-5p/SPAG9 axis to activate the JNK pathway, thus expediting HCC progression.

Prognostic biomarker identification and tumor classification in breast cancer patients by methylation and transcriptome analysis

Xiongdong Zhong et al.

Summary: In this study, 56 methylation-driven genes were identified and 10 prognostic genes were further screened using combined methylation and gene expression analysis. The protein levels of the 10 prognostic methylation-driven genes were detected by immunohistochemical experiments. A risk score signature was developed based on RNA-Seq data, and the risk score was confirmed to be an independent prognostic factor along with other clinical features.
