At the crossroads of immunotherapy for oncogene-addicted subsets of NSCLC

Summary: Immune-checkpoint inhibitors (ICI) have revolutionized the treatment of non-small-cell lung cancer (NSCLC), but identifying the minority of patients who benefit long-term remains challenging, especially among those with targetable oncogenic drivers who have been underrepresented in clinical trials. This review summarizes the associations between common oncogenic drivers of NSCLC and their sensitivity or resistance to ICIs, as well as the effects on the immune tumor microenvironment. It also highlights potential vulnerabilities that can be targeted to overcome primary resistance to ICIs caused by specific oncogenic drivers.

Hallmarks of Cancer: New Dimensions

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.
