3D Printing of Cell-Laden Microgel-Based Biphasic Bioink with Heterogeneous Microenvironment for Biomedical Applications

Summary: A generalizable strategy has been developed to formulate a cell-laden biphasic bioink for 3D extrusion bioprinting, which combines microgels with excellent rheological properties for printing and a hydrogel precursor for structural stability. This approach enables the printing of complex structures with high fidelity and offers great mechanical tunability and hyperelasticity, allowing for the encapsulation of different cell types for creating a heterogeneous cellular microenvironment at microscale. The MB bioink shows promise for various biomedical applications such as tissue engineering and soft robotics.

PNIPAAm microgels with defined network architecture as temperature sensors in optical stretchers

Summary: Stretching individual living cells with light is a common method for evaluating their mechanical properties. However, the heat generated by the laser light used in optical stretchers can unintentionally alter the cells' mechanical properties. To address this issue, researchers have developed cell-sized elastic poly(N-isopropylacrylamide) (PNIPAAm) microgels that can indicate temperature changes through hydrogel swelling. By analyzing the effects of different gelation methods on the microgel network heterogeneity, micromechanics, and temperature response, the researchers have enhanced the use of standardized microgels for calibrating and validating cell mechanics analysis.

3D Printing of Strong and Tough Double Network Granular Hydrogels

Summary: Many soft natural tissues possess unique mechanical properties achieved through complex interactions between structure and composition. To address the lack of control in soft synthetic materials, a novel 3D printing approach called double network granular hydrogels (DNGHs) has been introduced. These DNGHs are stiff enough to support high tensile loads and significantly tougher than pure polymeric networks. This ink allows for the printing of strong and tough objects with high shape fidelity, opening up new possibilities for soft robotic applications.
