Muller Glial Expression of REDD1 Is Required for Retinal Neurodegeneration and Visual Dysfunction in Diabetic Mice

William P. Miller et al.

Summary: Clinical studies support the role of REDD1 protein in ischemic retinal complications caused by diabetes. The study found that REDD1 expression specifically in Muller glia contributes to diabetes-induced retinal pathology, including gliosis, neurodegeneration, and impaired vision.

Clinical electroretinography in diabetic retinopathy: a review

Jason McAnany et al.

Summary: The electroretinogram (ERG) is a noninvasive technique that evaluates retinal function and has become increasingly important in studying diabetic retinopathy. Recent clinical studies using various types of ERG have provided evidence of neural dysfunction in diabetic patients, even in those without apparent retinal vascular abnormalities. The ERG has the potential to play a growing role in understanding the natural history of neural dysfunction in diabetes and as an outcome measure in clinical trials targeting neural preservation in diabetic retinopathy.

Morpho-functional analysis of the early changes induced in retinal ganglion cells by the onset of diabetic retinopathy: The effects of a neuroprotective strategy

Rosario Amato et al.

Summary: This study examines the early responses of retinal ganglion cells (RGCs) to diabetes-induced metabolic stress and the effects of the neuroprotective somatostatin analog octreotide (OCT). The results show that diabetes causes a decline in RGC function, but OCT can counteract this effect. Different subtypes of RGCs have varying responses to diabetes and OCT treatment. This study highlights the importance of neuronal protection in the early stages of diabetic retinopathy for maintaining RGC functionality.

Tribbles Homolog 3 Mediates the Development and Progression of Diabetic Retinopathy

Priyamvada M. Pitale et al.

Summary: TRIB3 plays a crucial role in diabetic retinopathy by controlling retinal glucose flux, reprogramming cellular metabolism, and regulating inflammatory gene expression. Inhibition of TRIB3 in mouse models shows significant effects, potentially making it a novel therapeutic target for diabetic retinopathy.

Effect of Intravitreous Anti-Vascular Endothelial Growth Factor vs Sham Treatment for Prevention of Vision-Threatening Complications of Diabetic Retinopathy The Protocol W Randomized Clinical Trial

Raj K. Maturi et al.

Summary: The study found that in eyes with moderate to severe NPDR, using aflibercept compared to sham treatment can reduce the proportion of eyes that develop proliferative diabetic retinopathy or non-center involved diabetic macular edema causing vision loss. However, preventive treatment did not confer visual acuity benefit compared with observation plus treatment with aflibercept only after development of PDR or vision-reducing CI-DME within two years.

Summary: The study aimed to evaluate the safety and efficacy of as-needed intravitreal aflibercept injections in managing diabetic retinopathy in eyes without diabetic macular edema. Results showed that most patients required re-treatment every 3-4 months, and deterioration of PLI appeared to precede DRSS level worsening.

Photoreceptor cells and RPE contribute to the development of diabetic retinopathy

Deoye Tonade et al.

Summary: Diabetic retinopathy is a leading cause of blindness, traditionally considered a vascular disease, but recent research has shown abnormalities in the neural retina. Evidence suggests that photoreceptors and retinal pigment epithelium in the outer retina play a key role in the development of vascular lesions in DR, presenting a novel therapeutic target for inhibiting the disease.
