Brain-Penetration and Neuron-Targeting DNA Nanoflowers Co-Delivering miR-124 and Rutin for Synergistic Therapy of Alzheimer's Disease

Summary: Alzheimer's disease (AD) is a common form of dementia in elderly individuals, with no effective treatment currently available. This study presents a novel DNA nanoflowers-based delivery system for AD therapy, which allows exogenous supplementation of miR-124 to ameliorate AD-like pathology. The nanosystem demonstrates promising results in increasing miR-124 levels in the brain and achieving robust therapeutic efficacy in a mouse model of AD.

Alteration of miRNAs in Small Neuron-Derived Extracellular Vesicles of Alzheimer's Disease Patients and the Effect of Extracellular Vesicles on Microglial Immune Responses

Summary: Alzheimer's disease (AD) is a severe neurodegenerative disease in the elderly population, and there is still a lack of definitive diagnosis and treatment methods. This study identified let-7e as a potential biomarker for AD diagnosis by studying the altered miRNA content in small neuron-derived extracellular vesicles (sNDEVs). Furthermore, it was found that AD patient-derived sNDEVs induce a neuroinflammatory response.

Summary: Alzheimer's disease (AD) is the most common neurodegenerative disease in older adults, and identifying peripheral blood-based biomarkers for AD diagnosis is urgently needed. By analyzing the miRNA profile in human serum exosomes, potential biomarkers, such as miR-30b-5p, miR-22-3p, and miR-378a-3p, were found to distinguish AD from healthy controls (HC). The combination of these three miRNAs showed improved diagnostic capability, indicating their potential as biomarkers for AD diagnosis.
