Impact of comorbidity scores and lifestyle factors in curative radiotherapy in laryngeal cancer

Zdenka Pechacova et al.

Summary: The study aimed to investigate the impact of comorbidities and lifestyle factors on treatment outcomes in laryngeal cancer patients. The results showed that baseline performance status, weight loss, anemia, associated comorbidities, history of alcohol abuse, and marital status were significant predictors of overall survival in patients treated with radio(chemo)therapy for laryngeal cancer.

Inhibiting the inhibitors: Development of the IAP inhibitor xevinapant for the treatment of locally advanced squamous cell carcinoma of the head and neck

Robert L. Ferris et al.

Summary: Standard of care for patients with locally advanced squamous cell carcinoma of the head and neck is surgery followed by chemoradiotherapy or definitive chemoradiotherapy. However, many patients experience disease recurrence or metastasis within 2 years, indicating a need for novel treatment options. Overexpression of inhibitor of apoptosis proteins (IAPs) has been identified as a key factor in treatment resistance and poor outcomes. Xevinapant, a potent IAP inhibitor, has shown promising results in combination with chemoradiotherapy in a phase 2 trial. This article explores the current treatment landscape and highlights the potential of targeting IAPs for improved outcomes.

Targeted therapy for head and neck cancer: signaling pathways and clinical studies

Qingfang Li et al.

Summary: Head and neck cancer is a malignant and genetically complex disease that is difficult to treat. It is the sixth most common cancer, with tobacco, alcohol, and human papillomavirus as major risk factors. HNC is heterogeneous and treatment remains challenging. Despite current therapies, recurrence and metastasis are common, and resistance to chemotherapy and targeted therapies is a problem. Therefore, it is urgent to explore more effective and tolerable targeted therapies to improve outcomes for HNC patients.

Xevinapant or placebo plus chemoradiotherapy in locally advanced squamous cell carcinoma of the head and neck: TrilynX phase III study design

Jean Bourhis et al.

Summary: Xevinapant is a new type of cancer treatment that enhances the effects of chemotherapy and radiotherapy. TrilynX is a randomized, double-blind, phase III study that aims to evaluate the efficacy of xevinapant in patients with locally advanced squamous cell carcinoma of the head and neck. The study's primary endpoint is event-free survival, and secondary endpoints include progression-free survival, locoregional control, overall survival, and safety.

Selective local postoperative radiotherapy for T3-T4 N0 laryngeal cancer

Meltem Dagdelen et al.

Summary: The appropriate postoperative radiotherapy dose and selective volume in T3-4 N0 laryngeal cancer patients treated with either total or partial laryngectomy were investigated. The findings suggest that local selective irradiation to the postoperative bed + stoma is enough in these patients.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Real-World Treatment Patterns and Outcomes in Patients with Head and Neck Cancer: Point-in-Time Survey of Oncologists in Italy and Spain

Prianka Singh et al.

Summary: A point-in-time survey of the management of platinum-eligible patients with recurrent or metastatic SCCHN in Italy and Spain revealed that despite following current European guidelines for first-line treatment, patients' quality of life remains poor. The primary site of the cancer in these patients was mainly in the pharynx, with 36% having metastatic disease. The most common treatments administered were the EXTREME or cetuximab-based regimens.

Apoptosis Deregulation and the Development of Cancer Multi-Drug Resistance

Christiana M. Neophytou et al.

Summary: This review discusses the deregulation of apoptotic pathways as a key factor leading to multidrug resistance (MDR) in cancer cells, along with the impact of the tumor microenvironment on apoptosis in MDR tumors. Therapeutic approaches targeting Bcl-2 family members, IAPs, and AKT, as well as the potential of epigenetic modifications in reversing MDR, are highlighted as important strategies against drug resistance.
