Summary: The involvement of mitochondria in neurodegenerative diseases is widespread. Recent development of bioactive molecules that modulate mitochondrial dynamics offers a new therapeutic approach. Activation of mitochondrial fusion and motility can improve mitochondrial pathology and symptoms in SOD1 mutant amyotrophic lateral sclerosis (ALS) patients and mice.

Mitochondrial Phenotypes in Genetically Diverse Neurodegenerative Diseases and Their Response to Mitofusin Activation

Summary: Mitochondrial fusion is crucial for mitochondrial and cellular health. Imbalance in mitochondrial fusion and fission, resulting in mitochondrial fragmentation, is observed in patients with genetic neurodegenerative diseases. By redirecting the metabolic pathway of patient fibroblasts, characteristic mitochondrial abnormalities and a specific gene expression profile can be induced. Pharmacological activation partially reverses these abnormalities, suggesting potential therapeutic targets for mitochondrial dysdynamism in various diseases.

Neuron-astrocyte transmitophagy is altered in Alzheimer's disease

Summary: This study found that the internalization and degradation of neuronal mitochondria by astrocytes is increased in Alzheimer's disease (AD), and this phenomenon has been confirmed in AD mice and human neurons. Summary: Recent research on amyotrophic lateral sclerosis (ALS) suggests that non-autonomous motor neuron death hypothesis may involve other cells besides motor neurons, with mitochondria playing a crucial role in ALS pathophysiology. Mutant SOD1 protein and down-regulation of OPA1 in the sympathoadrenal system of ALS mouse model lead to mitochondrial ultrastructure alterations and functional impairments prior to clinical symptoms. These findings highlight the potential therapeutic target of OPA1 and the relevance of sympathetic impairment in ALS progression.

Rational redesign based on pharmacophores generated a novel class of mitofusin activators, with analogue 5 incorporating a cycloalkyl linker group showing improved pharmacokinetic properties. Stereoisomeric studies revealed that the biological activity of analogue 5 resides in the trans-R/R configuration, 5B, which may have potential therapeutic implications for neurological conditions.
