Prelimbic Cortical Stimulation with L-methionine Enhances Cognition through Hippocampal DNA Methylation and Neuroplasticity Mechanisms

Chi Him Poon et al.

Summary: Declining DNA methylation and cognitive impairment are associated with normal aging. This study shows that DNA methylation may play a role in the efficacy of memory-enhancing therapies. Administration of PrL DBS and MET to aged animals resulted in enhanced hippocampal-dependent spatial memory, DNA methyltransferase-dependent methylation, upregulation of neuroplasticity-related genes, and inhibition of memory-suppressing gene calcineurin. The study also demonstrates the activation of the PKA-CaMKII alpha-BDNF pathway, increased hippocampal neurogenesis, and enhanced neurotransmission by MET-PrL DBS. Inhibition of DNA methyltransferase activity in young animals recapitulated behavioral deficits observed in aged animals and abolished the effects of PrL DBS.

Transcorneal electrical stimulation enhances cognitive functions in aged and 5XFAD mouse models

Wing Shan Yu et al.

Summary: Noninvasive visual stimulation is a novel concept that may be applicable for treating dementia. This study found that transcorneal electrical stimulation (TES) improved memory performance in aged mice and a mouse model of Alzheimer's disease, and reduced plaque deposition in the hippocampus of male 5XFAD mice. The findings suggest that TES shows promise as a potential treatment for cognitive dysfunction.

Distribution and inter-regional relationship of amyloid-beta plaque deposition in a 5xFAD mouse model of Alzheimer's disease

Ka Chun Tsui et al.

Summary: This study provides a comprehensive characterization of the distribution of Aβ plaques in the brain and their correlation with different brain regions. The results suggest a relationship between Aβ plaque deposition and the pathogenesis of Alzheimer's disease.

Chronic oral administration of adipoRon reverses cognitive impairments and ameliorates neuropathology in an Alzheimer's disease mouse model

Roy Chun-Laam Ng et al.

Summary: The decrease in circulating adiponectin levels is associated with neurodegeneration, neuroinflammation, and Alzheimer's disease pathologies. Increasing adiponectin through treatments like adipoRon can improve neuronal insulin signaling, memory functions, and reduce amyloid levels in Alzheimer's disease models.

Neurogenesis-dependent antidepressant-like activity of Hericium erinaceus in an animal model of depression

Pit Shan Chong et al.

Summary: Hericium erinaceus exerts antidepressant effects by promoting neurogenesis and reducing neuroinflammation through enhancing the BDNF-TrkB-CREB signaling pathway. The neurogenesis blocker temozolomide completely eliminates the antidepressant-like effects of HE, and HE induces anti-neuroinflammatory effects by decreasing astrocyte activation in the hippocampus.

Systematic phenotyping and characterization of the 5xFAD mouse model of Alzheimer's disease

Stefania Forner et al.

Summary: Mouse models of human diseases are crucial for studying pathogenic mechanisms and testing interventions. To effectively evaluate new therapeutic approaches, it is important to identify the most appropriate model and age. In this study, a detailed phenotypic characterization of the 5xFAD model was conducted to provide insights into age-related pathology.
