Blockade of beta-adrenergic receptors reduces cancer growth and enhances the response to anti-CTLA4 therapy by modulating the tumor microenvironment

Summary: The development of immune checkpoint inhibitors (ICI) has been an important breakthrough in cancer therapies, but only a limited number of patients benefit from them. Recently, the nonselective beta blocker propranolol has been shown to be effective in treating angiosarcoma and soft tissue sarcomas. It works by reducing tumor angiogenesis and promoting an anti-tumoral microenvironment with increased T cell infiltration. The combination of propranolol with immune checkpoint inhibitors can significantly enhance their efficacy.

Analysis of ctDNA in pet dogs can help address diagnostic needs in both veterinary and human oncology. This study demonstrates the feasibility of ctDNA analysis in dogs with hemangiosarcoma using a cost-effective approach. Further studies are needed to validate these findings and determine the role of ctDNA in assessing disease burden and treatment response in dogs with cancer.

Evaluation of a modified proportional margin approach for complete surgical excision of canine cutaneous mast cell tumours and its association with clinical outcome

Summary: A study on canine cutaneous mast cell tumours (MCTs) showed that a modified proportional margin approach with a 2 cm lateral margin upper limit was effective in surgically excising most MCTs, with no significant association between tumour size or grade and outcomes. Recurrence rate was 3% and post-operative metastases were seen in 4.6% of dogs. Overall, the technique was deemed suitable for guiding surgical excision of canine cutaneous MCTs.

Further research is needed to understand the causal and mechanistic connections of thrombocytopenia and its potential value in guiding treatment recommendations for affected dogs.

Combining propranolol with weak base chemotherapeutics could increase cytotoxicity and reduce the development of drug-resistant cell populations.
