LIN28 expression and function in medulloblastoma

Ahmed Maklad et al.

Summary: This study identified the expression of LIN28A and LIN28B in Medulloblastoma (MB) patient samples and cell lines. LIN28B expression was significantly upregulated in MB tissues compared to normal brain tissues and correlated with patient survival and metastasis rates. The results highlight the potential of LIN28B as a diagnostic and prognostic marker for MB and as a pharmacological target to suppress MB cell growth and stemness.

Pharmaco-proteogenomic profiling of pediatric diffuse midline glioma to inform future treatment strategies

Izac J. Findlay et al.

Summary: Diffuse midline glioma (DMG) is a deadly CNS tumor in children and adolescents, characterized by global hypomethylation of H3K27me3. Studies have shown high levels of intertumoral and intratumoral heterogeneity in DMG, which makes precision-based medicine challenging to execute successfully.

Identification of Let-7 miRNA Activity as a Prognostic Biomarker of SHH Medulloblastoma

Maximillian S. Westphal et al.

Summary: This study categorized medulloblastoma into molecular subgroups and found different survival rates among them. By analyzing the expression of molecular markers MYCN and let-7, one subgroup was further divided and it was found that high MYCN expression and high let-7 activity are associated with worse overall survival. These findings provide critical information for personalized treatment options for medulloblastoma.

The LIN28B/TGF-β/TGFBI feedback loop promotes cell migration and tumour initiation potential in cholangiocarcinoma

Nattapong Puthdee et al.

Summary: The study found that LIN28B is expressed in a subpopulation of CCA patients, promoting cancer stem cell-like properties such as enhanced cell migration and proliferation. TGF-beta-induced protein (TGFBI) was identified as a novel LIN28B target gene, suggesting a positive feedback loop between TGF-beta and LIN28B in promoting aggressive cell behavior during cholangiocarcinoma metastasis.

Hypomethylation-activated cancer-testis gene LIN28B promotes cell proliferation and metastasis in gastric cancer

Jiani Xu et al.

Summary: This study identified the CT gene LIN28B as being activated through hypomethylation in gastric cancer, affecting patient prognosis. It was found that accurate mediation of p methylation could activate LIN28B expression. Knockout of LIN28B suppressed proliferation, metastasis, and invasion of gastric cancer cells, while over-expression promoted metastasis and invasion.

Histone H3.3 K27M chromatin functions implicate a network of neurodevelopmental factors including ASCL1 and NEUROD1 in DIPG

Nichole A. Lewis et al.

Summary: The H3.3K27M mutation causes chromatin to adopt a more accessible configuration at regulatory regions for neurogenesis and NOTCH signaling genes in DIPG tumor cells, resulting in increased gene expression. This effect can be partially reversed by editing K27M back to the wild-type form.

Summary: By analyzing the single-cell transcriptomics, epigenomics, and spatial architectures of a comprehensive cohort of H3-K27M DMGs, this study reveals the heterogeneity of these mutations at different ages and midline locations. The findings also provide insights into the cell-intrinsic and -extrinsic features of these tumors in relation to age and anatomical location, which can be used for rational modeling and therapeutic interventions.

The KRAS/Lin28B axis maintains stemness of pancreatic cancer cells via the let-7i/TET3 pathway

Yawen Liu et al.

Summary: The study reveals that KRAS promotes Lin28B nuclear translocation through PKC beta, enhancing cellular migration and proliferation in pancreatic cancer. Nuclear Lin28B upregulates TET3 levels by blocking let-7i production, forming a feedback loop to maintain pancreatic cancer stemness. These findings highlight the important roles of Lin28B in KRAS-driven pancreatic cancer and offer implications for developing novel therapeutic strategies.

HMGA2 as a Critical Regulator in Cancer Development

Behzad Mansoori et al.

Summary: HMGA2 is a DNA architectural protein that regulates gene expression and is highly expressed in embryonic stem cells during development but re-expressed in most human malignancies, where it plays a critical role in promoting tumorigenesis through various mechanisms. HMGA2 increases cancer cell proliferation and resistance to chemotherapy, and influences DNA repair mechanisms and epithelial-to-mesenchymal transition, suggesting its potential as a diagnostic, prognostic, and therapeutic target in cancer.

LIN28B induces a differentiation program through CDX2 in colon cancer

Kensuke Suzuki et al.

Summary: The study uncovered the critical role of the LIN28B/CDX2 signaling axis in mediating colorectal cancer differentiation, with potential implications. Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.

LIN28B alters ribosomal dynamics to promote metastasis in MYCN-driven malignancy
