Myelination synchronizes cortical oscillations by consolidating parvalbumin-mediated phasic inhibition

Mohit Dubey et al.

Summary: PV+ interneurons in the brain are powerful inhibitors that regulate important cognitive processes. However, they have unusual, patchy myelination, and it is unclear how this affects their function. Loss of myelin in these interneurons reduces signal strength and increases the power of slow brain waves, as well as triggering epilepsy-like brain activity. Restoring myelination helps reverse these deficits and could be a potential therapeutic strategy for cognitive impairments.

Neonatal Oxidative Stress Impairs Cortical Synapse Formation and GABA Homeostasis in Parvalbumin-Expressing Interneurons

Till Scheuer et al.

Summary: Neonatal brain injury caused by preterm birth is often due to oxidative stress challenges. Studies suggest that increased oxygen exposure can lead to damage in GABAergic interneurons, which may explain the high incidence of psychiatric and behavioral alterations found in preterm infants.

Microglia-dependent remodeling of neuronal circuits

Joana R. Guedes et al.

Summary: Microglia are tissue-resident macrophages that play roles in surveillance, neuronal support, and immune defense in the brain. They participate in engulfing neuronal precursors, modulating synaptic signaling, and shaping synaptic plasticity. Microglia in different brain regions exhibit distinct characteristics that may potentially contribute to the maturation and function of specific neuronal circuits.

Deletion of Fmr1 in parvalbumin-expressing neurons results in dysregulated translation and selective behavioral deficits associated with fragile X syndrome

Magdalena Kalinowska et al.

Summary: This study found that de novo protein synthesis was elevated in hippocampal parvalbumin and somatostatin-expressing inhibitory neurons in Fmr1 knockout mice. Cell type-specific deletion of Fmr1 in parvalbumin-expressing neurons resulted in anxiety-like behavior, impaired social behavior, and dysregulated de novo protein synthesis. In contrast, deletion of Fmr1 in somatostatin-expressing neurons did not result in behavioral abnormalities and did not significantly impact de novo protein synthesis.

Parvalbumin interneuron vulnerability and brain disorders

Jacob B. Ruden et al.

Summary: PV-INs, as parvalbumin-expressing interneurons, are highly vulnerable to stressors and implicated in various neuro-psychiatric diseases. Factors contributing to their vulnerability include substantial energy requirements, strong excitatory drive, and a unique developmental trajectory. Understanding and addressing these stressors is essential for developing therapies to protect PV-INs and maintain critical brain functions.

Oxidative phosphorylation in creatine transporter deficiency

Shizhe Li et al.

Summary: X-linked creatine transporter deficiency (CTD) is a condition caused by pathogenic variants in the X-linked gene SLC6A8, leading to reduced phosphocreatine and total creatine concentrations. Despite the diminished role of creatine and phosphocreatine in oxidative phosphorylation in CTD, our study found no elevation of lactate or lowered pH, indicating that the brain still largely relies on oxidative metabolism for energy supply. Our results suggest that mitochondrial function could be a potential therapeutic target for CTD.

A new rat model of creatine transporter deficiency reveals behavioral disorder and altered brain metabolism

Lara Duran-Trio et al.

Summary: Creatine is crucial for tissues with high energy demand, and deficits in its synthesis or transport can lead to Cerebral Creatine Deficiency Syndromes. Among these syndromes, X-linked creatine transporter deficiency is the most prevalent with no effective treatment so far. The newly introduced Slc6a8(Y389C) KI rat model enriches the spectrum of CTD models and provides valuable insights into the pathology of this disorder, serving as the first animal model carrying a specific point mutation.

Early role for a Na+,K+-ATPase (ATP1A3) in brain development

Richard S. Smith et al.

Summary: Research shows that the ATP1A3 gene is highly expressed in a subset of excitatory neurons during fetal cortical development, and persists in inhibitory neurons, including parvalbumin interneurons, in early postnatal development. The heteromeric Na+,K+-ATPase pump complex can form specific alpha-beta isoform combinations in different cell types, such as alpha 3-beta 1 in excitatory neurons and alpha 3-beta 2 in inhibitory neurons, indicating a cell-type basis for ATP1A3-associated diseases in pre- and postnatal stages of human cortex development.
