Novel STING-targeted PET radiotracer for alert and therapeutic evaluation of acute lung injury

Duo Xu et al.

Summary: Acute lung injury (ALI) is a common clinical emergency characterized by pulmonary edema and diffuse lung infiltration caused by inflammation. This study developed a novel STING-targeted PET tracer, [18F]FBTA, which showed strong binding affinity to STING and could be used for PET imaging to detect early lung inflammation and evaluate the efficacy of drug therapy in ALI mice. Furthermore, [18F]FBTA was found to have better specificity and distribution compared to [18F]fluorodeoxyglucose ([18F]FDG).

Renewal of embryonic and neonatal-derived cardiac-resident macrophages in response to environmental cues abrogated their potential to promote cardiomyocyte proliferation via Jagged-1-Notch1

Rong Chen et al.

Summary: Cardiac-resident macrophages (CRMs) have important roles in cardiac homeostasis, function, and regeneration. This study investigated the changes of CRMs during cardiac ontogeny and their contribution to cardiac regeneration. The findings reveal the dynamic subsets of CRMs during development and their ability to regulate cardiomyocyte proliferation, highlighting their potential in cardiac repair and regeneration.

The disbalance of LRP1 and SIRPα by psychological stress dampens the clearance of tumor cells by macrophages

Yanping Wu et al.

Summary: This study reveals the relationship between chronic psychological stress and tumorigenesis, highlighting the impaired macrophage phagocytosis as a contributing factor. The stress hormone glucocorticoid is identified as a principal detrimental factor that disrupts the balance of eat me and don't eat me signal receptors. The findings provide mechanistic insight into psychological stress-evoked tumor susceptibility and suggest the potential therapeutic strategy of targeting the balance of LRP1/SIRP alpha axis for tumor treatment.

The anti-inflammatory feature of glucagon-like peptide-1 and its based diabetes drugsdTherapeutic potential exploration in lung injury

Juan Pang et al.

Summary: GLP-1 receptor agonists (GLP-1RAs) have been developed as therapeutic agents for type 2 diabetes (T2D) since 2005. GLP-1R is expressed not only in pancreatic islets but also in other organs, especially the lung. The controversy surrounding extra-pancreatic GLP-1R expression still needs to be resolved, and more reliable GLP-1R antibodies should be utilized for further research. The anti-inflammatory features of GLP-1RAs have shown promising results, but whether these features are directly mediated by GLP-1R expressed in immune cells remains controversial.

Scutellarin inhibits caspase-11 activation and pyroptosis in macrophages via regulating PKA signaling

Jiezhou Ye et al.

Summary: Scutellarin, a flavonoid from Erigeron breviscapus, acts as an inhibitor for caspase-11 activation in macrophages, reducing pyroptosis by inhibiting the release of caspase-11p26 and generation of GSDMD-NT. It also suppresses the activation of NLRP3 inflammasome and regulates the PKA signaling pathway.

Gastrin, via activation of PPARα, protects the kidney against hypertensive injury

Daqian Gu et al.

Summary: It was found that gastrin protects against hypertensive nephropathy by regulating renal tubule cell apoptosis, promoting macrophage efferocytosis, and reducing renal inflammation. Additionally, the nuclear translocation of CCKBR induced by gastrin may act as a transcription factor of PPAR-alpha, which represents a novel signaling pathway in renal fibrosis.

Gastrin exerts a protective effect against myocardial infarction via promoting angiogenesis

Jinjuan Fu et al.

Summary: In this study, administration of gastrin improved cardiac function in post-MI mice by reducing fibrosis, decreasing cardiomyocyte apoptosis, and increasing angiogenesis in the infarct border zone. In vitro results showed that gastrin activated the PI3K/Akt/VEGF signaling pathway, promoting migration and tube formation of human coronary artery endothelial cells. The protective effect of gastrin was mediated through the CCK2R receptor. These findings highlight the potential of gastrin as a therapeutic target candidate for MI.
