Peptide-drug conjugates (PDCs): a novel trend of research and development on targeted therapy, hype or hope?

Summary: Peptide-drug conjugates (PDCs) are the next generation of targeted therapeutics that have enhanced cellular permeability and improved drug selectivity. PDCs have been developed for cancer, COVID-19, and metabolic diseases, but face challenges in stability, bioactivity, and long development time. This review summarizes the components and functions of PDCs and discusses strategies for improving their permeability, targeting, and stability. The future direction of PDCs includes coupling with toxin or supramolecular nanostructures. Clinical trials and drug delivery modes for PDCs are also discussed.
