Evaluating the efficacy of a priming dose of cyclophosphamide prior to pembrolizumab to treat metastatic triple negative breast cancer

Summary: This study evaluated the efficacy of using a low dose of cyclophosphamide (Cy) to deplete regulatory T cells (T-regs) before initiating pembrolizumab in patients with triple negative breast cancer (TNBC). The results showed that Cy did not significantly decrease T-regs before pembrolizumab and there was a rapid recovery in T-regs after the first cycle of therapy. Baseline samples with increased B cell gene expression were associated with clinical response and immune-related toxicity (IRT).

Tertiary lymphoid structures generate and propagate anti-tumor antibody-producing cells in renal cell cancer

Summary: The presence of intratumoral tertiary lymphoid structures (TLS) is associated with positive clinical outcomes and responses to immunotherapy in renal cell carcinoma (RCC). B cells are enriched in TLS and demonstrate diverse maturation stages, with the presence of fully mature clonotypes. Additionally, IgG-and IgA-producing plasma cells (PCs) disseminate into the tumor beds along fibroblastic tracks in TLS+ tumors, indicating potential anti-tumor effector activity. Furthermore, the presence of IgG-stained tumor cells correlates with therapeutic responses and progression-free survival in RCC patients treated with immune checkpoint inhibitors.

Summary: This prospective study aimed to evaluate the association between autoantibodies in the serum of cancer patients and immune-checkpoint inhibitor (ICI) therapy response and immune-related adverse events (irAEs). The results showed that there was no significant association between the presence of autoantibodies and treatment efficacy or higher grade irAEs, neither at treatment start nor after 8-12 weeks of ICI treatment.
