Nanoflow LC-MS Method Allowing In-Depth Characterization of Natural Heterogeneity of Complex Bacterial Lipopolysaccharides

Summary: The variable modification of outer membrane lipopolysaccharide (LPS) in Gram-negative bacteria contributes to bacterial pathogenesis. A new method using nanoflow reversed-phase high-performance liquid chromatography coupled with electrospray ionization Fourier transform mass spectrometry was developed for the analysis of complex bacterial LPS mixtures. This method provided detailed information on the composition of oligo/polysaccharides and lipid A domains of LPS species, showing high-resolution mass spectra of intact LPS.
