Structure, substrate recognition and initiation of hyaluronan synthase

Finn P. Maloney et al.

Summary: In this study, we investigated the substrate binding and polymer synthesis of hyaluronan synthase (HAS) using cryo-electron microscopy structures, biochemical analyses, and molecular dynamics simulations. Our findings provide detailed insights into the transmembrane process of HAS, including substrate selection, polymerization, and regulation of hyaluronan synthesis. This research is important for understanding the biosynthesis of acidic extracellular polysaccharides and one of the most abundant glycosaminoglycans in the human body.

Chemical editing of proteoglycan architecture

Timothy R. O'Leary et al.

Summary: Proteoglycans are important glycoconjugates that play a role in many cellular processes. However, the study of their structure and function has mainly focused on the glycosaminoglycan chains, overlooking other important elements such as core proteins and membrane localization. In this study, we present a comprehensive approach to investigate proteoglycan structure and biology by creating tailored semisynthetic modular proteoglycans that can be displayed on the cell surface. By using proteoglycan core proteins with unnatural amino acids, we were able to systematically study the binding and function of various proteoglycan-binding proteins.

Summary: Glycosaminoglycans (GAGs) play a role in the treatment of many human diseases, especially in thrombosis, due to their anticoagulant activity. Novel electrophoresis-mass spectrometry methods have been developed to directly elucidate the structures of larger GAG chains with more biological importance, aiding in understanding the structure-activity relationship and quality control of GAG drugs.

Design of Polymeric Carriers for Intracellular Peptide Delivery in Oncology Applications

Shixian Lv et al.

Summary: Peptides as promising cancer therapeutics have shown robust antitumor efficacy, but the translation into clinical therapies is hindered by inherent limitations in their structure, hindering intracellular delivery. Strategies to engineer polymeric materials for increased peptide delivery efficiency, especially cytosolic delivery, play a crucial role in potentiating peptide-based cancer therapies. Future opportunities for peptides in cancer treatment lie in the design of polymer nanocarriers for optimized peptide delivery.
