A SWELL time to develop the molecular pharmacology of the volume-regulated anion channel (VRAC)

Eric E. Figueroa et al.

Summary: The newly emerging roles of LRRC8 volume-regulated anion channels (VRAC) in the treatment of epilepsy, type 2 diabetes, and other human diseases raise important questions about their therapeutic potential. However, the lack of potent and specific small-molecule inhibitors and activators of VRAC hinders the evaluation of VRAC as a viable drug target. Recent progress has been made in developing the molecular pharmacology of VRAC through screening FDA-approved drugs, leading to the discovery of novel VRAC inhibitors and activators. This sets the stage for further research on the molecular pharmacology and therapeutic potential of VRAC.

LRRC8A is dispensable for a variety of microglial functions and response to acute stroke

James R. Cook et al.

Summary: Microglia function does not critically rely on VRAC, although VRAC plays an important role in cell volume regulation. It does not affect phagocytic activity, cell migration, or P2YR12-dependent chemotaxis. Loss of VRAC in microglia does not impact microglial morphology or the extent of ischemic damage. VRAC may be targetable in other CNS cell types.

VI-116, A Novel Potent Inhibitor of VRAC with Minimal Effect on ANO1

Dongkyu Jeon et al.

Summary: Volume-regulated anion channel (VRAC) plays a crucial role in cell volume regulation, and the lack of potent and selective VRAC inhibitors has limited the research in this field. In this study, a novel VRAC inhibitor, VI-116, was identified, which showed high selectivity for VRAC without affecting intracellular calcium signaling and hERG K+ channel activity.

Sphingosine-1-phosphate induces migration of microglial cells via activation of volume-sensitive anion channels, ATP secretion and activation of purinergic receptors

Danyal Zahiri et al.

Summary: The study demonstrated that S1P induces anion channel activity in microglial BV-2 cells via VRAC, leading to cell migration and increased ATP levels. The involvement of S1P receptor 1 and G proteins plays a crucial role in this process.

On the molecular nature of large-pore channels

Johanna Syrjanen et al.

Summary: Membrane transport plays a crucial role in controlling cellular processes, with large-pore channels serving as important conduits for ions and metabolites. These channels are formed by different protein families such as connexin, innexin, and pannexin, each contributing to diverse cellular functions.
