Density Functional Theory Investigation of the Biocatalytic Mechanisms of pH-Driven Biomimetic Behavior in CeO2

Hongyang Ma et al.

Summary: There is significant interest in the pH-dependent properties of cerium oxide nanoparticles, which exhibit antioxidant activity at basic pH but cytotoxic prooxidant activity at acidic cancer cell pH. This study investigates the atomic-scale behavior of CeO2 nanoparticles with oxygen vacancies under different pH conditions using density functional theory calculations. The results suggest that pH plays a critical role in the interaction between reactive oxygen species and the defective CeO2 surface, leading to either antioxidant or prooxidant activity.

Superoxide dismutase-loaded porous polymersomes as highly efficient antioxidant nanoparticles targeting synovium for osteoarthritis therapy

Tao Gui et al.

Summary: Oxidative stress and reactive oxygen species play important roles in the development and progression of osteoarthritis (OA). This study utilized SOD-loaded nanoparticles to deliver antioxidant enzymes to mouse knee joints, demonstrating the accumulation of nanoparticles in the synovium and reduction of oxidative damage. Intra-articular injections of the nanoparticles were effective in attenuating OA initiation and progression, via reduction of reactive oxygen species production and catabolic protease synthesis.

Pentraxin 3 regulated by miR-224-5p modulates macrophage reprogramming and exacerbates osteoarthritis associated synovitis by targeting CD32

Jianbin Yin et al.

Summary: An imbalance in M1/M2 macrophage polarization plays a crucial role in the progression of osteoarthritis (OA). Pentraxin 3 (PTX3) is found to be highly expressed in OA M1-polarized macrophages and is involved in the occurrence and development of OA. PTX3 primarily affects OA through macrophages and the paracrine interaction between macrophages and chondrocytes produces a feedback loop that enhances synovitis and cartilage damage. This study identifies a functional pathway important to OA development and suggests that blocking this pathway and PTX3 may prevent and treat OA.

Mechanisms and therapeutic implications of cellular senescence in osteoarthritis

Philip R. Coryell et al.

Summary: The development of osteoarthritis (OA) is correlated with an increase in senescent cells in joint tissues, with senescence-associated secretory phenotype (SASP) implicated in cartilage degradation and OA. Developing therapeutics like senolytics or senomorphics to eliminate or alter senescent cells may halt the progression and pathogenesis of OA.

Amelioration of post-traumatic osteoarthritis via nanoparticle depots delivering small interfering RNA to damaged cartilage

Sean K. Bedingfield et al.

Summary: In this study, intra-articular injection of nanoparticles targeting collagen and delivering small interfering RNA for matrix metalloproteinase 13 effectively protected cartilage integrity and reduced disease progression in mice with osteoarthritis. The inhibition of MMP13 suppressed genes associated with tissue restructuring, angiogenesis, innate immune responses, and proteolysis. Targeting collagen in damaged extracellular matrix of arthritic cartilage may be an effective strategy for treating osteoarthritis.
