Summary: Cell signalling is tightly regulated by specialized membranes and vesicles containing unique lipids and proteins. Paladin, identified as a PI(4,5)P-2 phosphatase, regulates VEGFR2 endosomal signalling and angiogenesis by interacting with VEGFR2 in endosomal and Golgi compartments. Inhibition of Paladin could potentially modulate VEGFR2 signalling and angiogenesis without directly inhibiting the receptor.

EVL regulates VEGF receptor-2 internalization and signaling in developmental angiogenesis

Summary: Endothelial tip cells are essential for VEGF-induced angiogenesis, and Ena/VASP proteins, particularly EVL, play a pivotal role in this process. EVL deletion compromises the radial sprouting of the vascular plexus and reduces endothelial tip cell density and filopodia formation, impairing VEGF-induced endothelial cell proliferation and sprouting. This study demonstrates that endothelial EVL regulates sprouting angiogenesis via VEGF receptor-2 internalization and signaling through the MAPK/ERK pathway.
