Signal Peptide Features Determining the Substrate Specificities of Targeting and Translocation Components in Human ER Protein Import

Summary: Approximately 30% of polypeptides enter the secretory pathway in human cells, and this process involves various signal peptides and transmembrane helices. Most precursor polypeptides are integrated into or translocated across the ER membrane via the Sec61 channel, but certain precursor polypeptides require specific auxiliary components for gating. Recent studies have provided insights into the client spectrum and specificities of some auxiliary components. By combining siRNA-mediated depletion with quantitative proteomics, we can determine the preference of certain pathways or components in intact human cells.

Utilizing TRAP and Sec62/Sec63 complexes as models, the study aimed to detect interactions of membrane proteins in living mammalian cells under physiological conditions, revealing multiple protein-protein interactions of diverse topological layouts.

The translocon-associated protein (TRAP) complex regulates quality control of N-linked glycosylation during ER stress

Summary: This study demonstrates that cells lacking the SSR3 or SSR4 subunits can restore N-glycosylation over time and reduce the transcriptional signature of ER stress. The protein levels of the SSR3 subunit are dependent on ER stress and UBE2J1, coordinating upstream N-glycosylation proficiency with downstream ER-associated degradation and proteostasis. The TRAP complex plays a regulatory role in maintaining ER glycoprotein quality control under stress conditions in both non-transformed and tumor cells.
