Nuts and bolts of the salt-inducible kinases (SIKs)

Nicola J. Darling et al.

Summary: The salt-inducible kinases (SIKs) are closely related to AMP-activated protein kinase (AMPK) and require phosphorylation by LKB1 for activation. They are phosphorylated by PKA to be inactivated. The main substrates of SIKs are CRTCs and Class 2a HDACs, and their regulation by SIKs influences various physiological processes. The reversible regulation of these substrates by SIKs highlights their potential in therapeutic interventions for diseases.

An NKX2-1/ERK/WNT feedback loop modulates gastric identity and response to targeted therapy in lung adenocarcinoma

Rediet Zewdu et al.

Summary: Cancer cells can switch lineage during progression and in response to therapy, with NKX2-1 loss leading to a specific subtype of lung cancer. This loss impacts the spectrum of driver oncogenes and response to inhibitors, suggesting a complex relationship between lineage specifiers and oncogenic pathways in lung adenocarcinoma regulation.

The PEMDAC phase 2 study of pembrolizumab and entinostat in patients with metastatic uveal melanoma

Lars Ny et al.

Summary: The combination of HDAC inhibitor entinostat with anti-PD-1 antibody pembrolizumab shows durable responses in a subset of patients with metastatic uveal melanoma. Factors such as wildtype BAP1 gene, low tumor burden, and iris melanoma may be associated with better treatment response and longer survival.

Entinostat induces antitumor immune responses through immune editing of tumor neoantigens

Andrew S. Truong et al.

Summary: The study highlights the immunomodulatory effects of entinostat on tumor neoantigens, remodeling the tumor immune microenvironment, leading to a strong antitumor response with increased neoantigen-specific T cells. Combination treatment with entinostat and PD-1 blockade results in complete responses and long-term immune memory, providing a strong preclinical rationale.
