Structure of monkeypox virus DNA polymerase holoenzyme

Qi Peng et al.

Summary: The World Health Organization declared monkeypox a global public health emergency in July 2022 and urgent measures are needed. Using cryo-electron microscopy, the structure of the monkeypox virus polymerase holoenzyme in complex with DNA was determined. It was found that the holoenzyme utilizes a forward sliding clamp processivity mechanism for viral DNA replication, similar to other B-family DNA polymerases. These findings provide insight into the replication mechanism of the monkeypox virus and may aid in the development of anti-poxvirus drugs.

Summary: This review addresses the unique challenges of replicating a 195-kb DNA genome in the cytoplasm, including the release and deposition of the genome, reorganization of the endoplasmic reticulum to form a replication factory, tailoring of the cellular milieu to support high-fidelity replication, and encapsidation of newly synthesized genomes.

Phylogenomic characterization and signs of microevolution in the 2022 multi-country outbreak of monkeypox virus

Joana Isidro et al.

Summary: In this study, shotgun metagenomics were used to rapidly reconstruct and characterize the genome sequences of the largest monkeypox virus outbreak in non-endemic countries so far. The findings indicate that the outbreak most likely has a single origin and that the virus belongs to clade 3. Furthermore, the study suggests the involvement of host APOBEC3 in viral evolution and reveals signs of ongoing microevolution in the virus.

Monkeypox and Its Possible Sexual Transmission: Where Are We Now with Its Evidence?

Ranjit Sah et al.

Summary: Monkeypox is a rare disease that has been increasing in incidence in different countries. It is primarily transmitted through zoonosis in endemic areas and through human-to-human transmission in non-endemic regions. Symptoms include flu-like symptoms, rash, and sexual contact is a major mode of transmission.

Monkeypox: disease epidemiology, host immunity and clinical interventions

Fok-Moon Lum et al.

Summary: In this review, the clinical, epidemiological, and immunological aspects of monkeypox virus (MPXV) infections are examined, with a focus on host immunity mechanisms. The unique epidemiological and pathological characteristics of the current non-endemic outbreak of the virus are also considered, along with vaccines, therapeutics, and outstanding research questions.

Solution Structure of the C-terminal Domain of A20, the Missing Brick for the Characterization of the Interface between Vaccinia Virus DNA Polymerase and its Processivity Factor

Beate Bersch et al.

Summary: Poxviruses are enveloped viruses with a linear, double-stranded DNA genome, and viral DNA synthesis is achieved by a DNA polymerase holoenzyme composed of three essential proteins. The catalytic subunit E9 forms a complex with the heterodimeric processivity factor made up of D4 and A20, which have a specific interaction involving a poxvirus-specific insertion. The interface between E9 and A20 comprises conserved hydrophobic residues and has been structurally characterized, providing insights into their molecular interactions.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
