DFT and CBS Study of Ethyl Acetate Conformers in the Neutral Hydrolysis

Drug repurposing for Alzheimer's disease: in silico and in vitro investigation of FDA-approved drugs as acetylcholinesterase inhibitors

Summary: This study used computational virtual screening to identify potential drugs for the treatment of Alzheimer's disease, including antidiabetic and antimalarial drugs. Experimental results showed that these drugs had inhibitory effects on acetylcholinesterase. It was also found that antifungal drugs might have the potential to be AChE inhibitors. Molecular dynamics simulations provided insights into the interactions and stability of these potential drugs with proteins.

Design, synthesis, molecular modeling and neuroprotective effects of a new framework of cholinesterase inhibitors for Alzheimer's disease

Summary: A new series of compounds were designed, synthesized, and evaluated as potential neuroprotective agents against Alzheimer's disease. These compounds showed inhibitory activity against AChE and BChE and were found to be selective and non-cytotoxic. Kinetic and molecular modeling studies indicated that they could act as dual-site inhibitors for both enzymes and also exhibited promising anti-inflammatory activity.

Decreased pH in the aging brain and Alzheimer's disease

Summary: The study compared pH levels in the human brain and cerebrospinal fluid (CSF) in postmortem control and Alzheimer's disease cases using publicly available data sets. Results showed a reduction in pH with aging and a further decrease in Alzheimer's disease cases. In vivo experiments demonstrated that low pH CSF infusion increased amyloid beta plaque load, while in vitro studies showed that mild acidosis reduced inflammation and amyloid beta uptake by microglia. These findings suggest that brain acidosis may play a role in Alzheimer's disease pathophysiology.
