Time Rules the Efficacy of Immune Checkpoint Inhibitors in Photodynamic Therapy

Qinghua Wu et al.

Summary: This study investigates the timing of intervention with immune checkpoint inhibitors (ICI) after photodynamic therapy (PDT) and its impact on tumor efficacy. A tumor-targeting nanomedicine encapsulating indocyanine green is prepared, and ICI is administered 5 days after PDT. The optimal combination significantly suppresses tumor growth and eliminates metastasis. This research provides theoretical guidance for enhancing immune checkpoint blockade therapy (ICBT) through intervention timing.

Combination of a novel heat shock protein 90-targeted photodynamic therapy with PD-1/PD-L1 blockade induces potent systemic antitumor efficacy and abscopal effect against breast cancers

Kensuke Kaneko et al.

Summary: Combining HS201-PDT with anti-PD-L1 antibody showed enhanced antitumor activity, upregulated CXCR3 gene signature, and improved tumor infiltration by T cells, suggesting the potential of HS201-PDT combined with PD-1/PD-L1 blockade as a treatment strategy for breast cancer. Receptor-ligand interactome analysis revealed enhanced signaling pathways associated with CD8+T cell activation in the combination group.

Combinatorial therapeutic approaches of photodynamic therapy and immune checkpoint blockade for colon cancer treatment

Yang Hao et al.

Summary: Photodynamic therapy (PDT) using indocyanine green (ICG) as a photosensitizer shows promising results in cancer treatment by generating reactive oxygen species (ROS) and inducing immune responses. This study demonstrates that ICG enhances the cell-killing effects of PDT and induces immunogenic cell death (ICD). Combined with immune checkpoint blockade, PDT under near-infrared (NIR) illumination prolongs the survival rate in colorectal tumor-bearing mice by inducing a series of immune responses.

In situ immunogenic clearance induced by a combination of photodynamic therapy and rho-kinase inhibition sensitizes immune checkpoint blockade response to elicit systemic antitumor immunity against intraocular melanoma and its metastasis

Seohyun Kim et al.

Summary: A targeted immunotherapeutic strategy involving local photodynamic therapy, a rho-kinase inhibitor, and PD-1/PD-L1 immune checkpoint blockade was proposed for uveal melanoma. This treatment approach demonstrated efficacy in promoting immunogenic clearance of primary tumors, preventing metastasis, and improving survival outcomes for patients.

Photodynamic therapy synergizes with PD-L1 checkpoint blockade for immunotherapy of CRC by multifunctional nanoparticles

Zeting Yuan et al.

Summary: Recent studies show that photodynamic therapy (PDT) can enhance the therapeutic effects of PD-L1 blockade in colorectal cancer (CRC) by inducing tumor cell death, stimulating immune response, and preventing tumor recurrence through upregulation of PD-L1 expression via the hypoxia-inducible factor 1a (HIF-1a) signaling pathway. This combination therapy using multifunctional nanoparticles loaded with photosensitized mTHPC (mTHPC@VeC/T-RGD NPs) shows promise in improving the response rate of anti-PD-L1 checkpoint blockade immunotherapies in CRC.
