Therapeutic Exon Skipping Through a CRISPR-Guided Cytidine Deaminase Rescues Dystrophic Cardiomyopathy in Vivo

Jia Li et al.

Summary: The study analyzed a novel murine model of DMD and examined the feasibility of using a cytidine base editor for exon skipping in vivo. The results showed that AAV9-eTAM treatment in the mouse model restored dystrophin and improved cardiac and skeletal muscle functions significantly.

Precise correction of Duchenne muscular dystrophy exon deletion mutations by base and prime editing

F. Chemello et al.

Summary: This study demonstrated the use of adenine base editor to restore dystrophin expression in cardiomyocytes affected by Duchenne muscular dystrophy mutation, as well as reframing the dystrophin open reading frame. In vivo experiments with mice showed successful gene editing and disease correction, indicating the potential of nucleotide editing for diverse DMD mutations with minimal genome modification.

In vivo somatic cell base editing and prime editing

Gregory A. Newby et al.

Summary: Recent advances in genome editing technologies have made single-dose cures for genetic diseases more promising. Base editors and prime editors offer greater precision and efficiency compared to nuclease-dependent editing approaches, with less unwanted byproducts. Delivery of these editors in animal models has shown promise for in vivo therapeutic genome editing.

Efficient precise in vivo base editing in adult dystrophic mice

Li Xu et al.

Summary: Recent advances in base editing have allowed for precise correction of disease-causing mutations, but challenges remain in terms of the size of base editors and their off-target activities, as well as the requirement for a specific protospacer adjacent motif (PAM) nearby the mutation site. This study successfully modified an NG-targeting adenine base editor to overcome these challenges and demonstrated its high efficiency in editing a Duchenne muscular dystrophy (DMD) mutation in adult mice, showing promising results with systemic delivery. The authors' approach shows potential for the treatment of monogenic diseases using base editing techniques, addressing limitations in targeting feasibility and packaging capacity.

Intramyocardial Transplantation of Human iPS Cell-Derived Cardiac Spheroids Improves Cardiac Function in Heart Failure Animals

Shinji Kawaguchi et al.

Summary: The severe shortage of donor hearts hampers cardiac transplantation, leading to high expectations for cardiac regenerative therapies. hiPSC-derived cardiomyocytes offer a realistic cell source for heart recovery. Avoiding teratoma formation and scaling up cardiomyocyte culture are essential for clinical applications. Spheroids and gelatin hydrogel combination has shown success in recovering reduced ejection fraction, and a feasible transplantation strategy for regenerative cardiomyocytes has been established in this study.
