Proteomic evaluation of human osteoblast responses to titanium implants over time

Francisco Romero-Gavilan et al.

Summary: This study investigated the effect of sandblasted acid-etched (SAE) titanium implants on the biological responses of human osteoblasts using proteomic analysis. The results showed that SAE-Ti increased mineralization and expression of specific genes in osteoblasts, thereby enhancing osseointegration.

Modulating Osteoimmune Responses by Mesoporous Silica Nanoparticles

Sepanta Hosseinpour et al.

Summary: This Review summarizes the osteoimmunomodulation of mesoporous silica nanoparticles (MSNs) in bone regeneration. The effects of MSN characteristics, including surface topography and charge, particle size, and ion release, on immune cells and osteogenesis were discussed. Appropriate doses of MSNs can inhibit inflammation and create an immune microenvironment beneficial for bone regeneration by activating immune cells and stimulating cytokine release. Further research is needed to explore and clarify the underlying mechanisms, including crosstalk between various types of immune cells and how to design MSNs to create a suitable immune environment for osteogenesis.

Substrate-dependent modulation of the leukotriene A4 hydrolase aminopeptidase activity and effect in a murine model of acute lung inflammation

Kyung Hyeon Lee et al.

Summary: The aminopeptidase activity of leukotriene A(4) hydrolase has become a therapeutic target for modulating host immunity. The modulator 4-methoxydiphenylmethane (4MDM) can inhibit PGP hydrolysis and LTA(4)H aminopeptidase activity, but activate the same enzyme target with alternative substrates. In a murine model, 4MDM was found to modulate the host neutrophilic response independent of clearing PGP.

Calcium silicate enhances immunosuppressive function of MSCs to indirectly modulate the polarization of macrophages

Haiyan Li et al.

Summary: This study found that the ionic products of calcium silicate (CS), a representative BSCs, can enhance the immunosuppressive function of human bone marrow mesenchymal stem cells (HBMSCs) by up-regulating the expression of immunosuppressive factors via NF-kappa B pathway. Additionally, CS-activated HBMSCs have stronger stimulatory effects on M2 polarization of macrophages and early osteogenic differentiation of HBMSCs compared to CS ionic products, providing insights into the mechanism of BSCs enhancing bone regeneration and the importance of considering the effects of biomaterials on the immunomodulatory function of tissue forming cells.
