Liang-Zhong Niu et al.

Summary: CG and CHG methylation levels in the rapid shoot growth stages of woody bamboos were significantly decreased, while CHH methylation was strongly associated with shoot developmental time or age. DNA methylation plays a critical role in regulating the bamboo rapid shoot growth, and CHH methylation is accumulated in transposable element regions.

5-Azacytidine increases tanshinone production in Salvia miltiorrhiza hairy roots through epigenetic modulation

Bo-Cheng Yang et al.

Summary: Recent studies have found strong connections between epigenetic modulation and secondary metabolites in plants. In this study, the effects of the DNA methylation inhibitor 5-azacytidine (5-Az) on secondary metabolite production, gene expression, and DNA methylation levels were investigated in the hairy roots of the medicinal plant Salvia miltiorrhiza. Results showed that the content of tanshinones, a type of secondary metabolite, increased by 1.5-5 times in the hairy roots treated with 5-Az. The methylation pattern in the promoter region of the gene encoding copalyl diphosphate synthase (CPS) was altered during 5-Az treatment, and several transcription factor binding sites were identified. This study provides evidence for a possible mechanism of DNA methylation in regulating tanshinone biosynthesis in the hairy roots of S. miltiorrhiza.

A 2-oxoglutarate-dependent dioxygenase converts dihydrofuran to furan in Salvia diterpenoids

Jiao-Jiao Song et al.

Summary: A 2-oxoglutarate-dependent dioxygenase (2-ODD) gene responsible for tanshinone IIA biosynthesis in Salvia miltiorrhiza was identified in this study. The gene, known as Sm2-ODD14, acts as a dehydrogenase and catalyzes the aromatization of the furan ring. The study also showed that Sm2-ODD14 controls the metabolite flux from dihydrofuran- to furan-tanshinones, which impacts the medicinal properties of S. miltiorrhiza.
