Developmental Programming: Prenatal Testosterone Excess on Liver and Muscle Coding and Noncoding RNA in Female Sheep

Nadia Saadat et al.

Summary: This study investigates the transcriptional changes and transcriptome signature of prenatal testosterone excess-induced hepatic and muscle-specific metabolic disruptions. The findings suggest that downregulation of mitochondria, lipid catabolism, and PPAR signaling genes in the liver and dysregulation of mitochondrial and ncRNA gene pathways in muscle contribute to lipotoxic and insulin-resistant hepatic and muscle phenotype.

Analysis of Upstream Regulators, Networks, and Pathways Associated With the Expression Patterns of Polycystic Ovary Syndrome Candidate Genes During Fetal Ovary Development

Rafiatu Azumah et al.

Summary: This study analyzed the expression of polycystic ovary syndrome (PCOS) candidate genes in bovine fetal ovaries and identified different expression patterns and upstream regulators associated with these genes. The early cluster of genes was mainly involved in mitochondrial function and oxidative phosphorylation, while the late cluster of genes was associated with stromal expansion, cholesterol biosynthesis, and steroidogenesis. These findings provide insights into the development and origins of PCOS, suggesting that multiple etiological pathways may contribute to the development of the syndrome.

Thada Is Dispensable for Female Fertility in Mice

Shan Han et al.

Summary: This study investigated the impacts of Thada gene deletion on ovarian function and reproductive outcomes using a mouse model. The results showed that Thada deletion did not significantly affect ovarian function and female fertility. Additionally, a high-fat-high-sugar diet did not alter this impact.

Hepatic Mitochondrial Dysfunction and Risk of Liver Disease in an Ovine Model of PCOS Males

Katarzyna J. Siemienowicz et al.

Summary: This study investigated the effects of prenatal androgenic overexposure on hepatic mitochondrial function and lipid metabolism in adolescent male sheep using an ovine model. The results suggest that excess in utero androgen exposure leads to a PCOS-like metabolic phenotype in male sheep, characterized by increased hepatic cholesterol and glycogen accumulation, dysregulated glucose and fatty acid metabolism, mitochondrial dysfunction, and signs of early liver fibrosis.

Daughters of polycystic ovary syndrome pregnancies and androgen levels in puberty: a Meta-analysis

Georgios Valsamakis et al.

Summary: The purpose of this study was to provide an overview and critical analysis of the literature on the circulating androgen levels in prepubertal and pubertal girls who have not been diagnosed with PCOS or precocious puberty, but have mothers with PCOS. The results showed that pubertal daughters of PCOS mothers had significantly higher levels of total testosterone and 17-OHP, and lower levels of SHBG, while prepubertal daughters of PCOS mothers had higher levels of SHBG compared to controls. These findings suggest that early intervention during puberty may be crucial in the development of the disease.

HMGA2 promotes breast cancer metastasis by modulating Hippo-YAP signaling pathway

Summary: This study found that HMGA2 is highly expressed in TNBC cells, and downregulation of HMGA2 inhibits the migration and invasion of TNBC, as well as tumor metastasis in vivo, through suppression of EMT and the Hippo-YAP pathway.

Summary: PCOS is a common complex disease in women with a strong genetic component, and studies have identified 14 genes potentially associated with the condition. These genes may play a role in mediating disease risk, with some already implicated in the hypothalamic-pituitary-gonadal signaling pathway.

HMGA2 as a Critical Regulator in Cancer Development

Behzad Mansoori et al.

Summary: HMGA2 is a DNA architectural protein that regulates gene expression and is highly expressed in embryonic stem cells during development but re-expressed in most human malignancies, where it plays a critical role in promoting tumorigenesis through various mechanisms. HMGA2 increases cancer cell proliferation and resistance to chemotherapy, and influences DNA repair mechanisms and epithelial-to-mesenchymal transition, suggesting its potential as a diagnostic, prognostic, and therapeutic target in cancer.

Glucocorticoid signaling and lipid metabolism disturbances in the liver of rats treated with 5α-dihydrotestosterone in an animal model of polycystic ovary syndrome

Danijela Vojnovic Milutinovic et al.

Summary: The study aimed to investigate the association between alterations in hepatic glucocorticoid signaling and disturbances of glucose and lipid metabolism in an animal model of PCOS. The results showed that DHT treatment increased corticosterone availability in the liver, leading to enhanced glucocorticoid receptor activation, stimulated triglyceride synthesis, and hypertriglyceridemia, without affecting gluconeogenesis and de novo lipogenesis.
