GPR43 activation-mediated lipotoxicity contributes to podocyte injury in diabetic nephropathy by modulating the ERK/EGR1 pathway

Jian Lu et al.

Summary: The activation of GPR43 contributes to podocyte injury in diabetic nephropathy by mediating lipotoxicity and modulating the ERK/EGR1 pathway. The study also found that GPR43 gene knockout inhibited cholesterol accumulation and autophagy inhibition. These findings suggest that GPR43 could be a potential target for treating podocyte injury in diabetic nephropathy.

Hydroxychloroquine alleviates renal interstitial fibrosis by inhibiting the PI3K/Akt signaling pathway

Dengren Li et al.

Summary: This study found that hydroxychloroquine (HCQ) can alleviate inflammation and cell apoptosis caused by renal fibrosis by inhibiting the activation of the PI3K/AKT and NF-kB pathways, improving the epithelial-mesenchymal transition (EMT) and extracellular matrix (ECM) deposition in renal tubular epithelial cells. This provides theoretical evidence for the treatment of renal fibrosis with HCQ.

Upregulation of TIPE1 in tubular epithelial cell aggravates diabetic nephropathy by disrupting PHB2 mediated mitophagy

Lei Liu et al.

Summary: This study investigated the role of TIPE1 in mitochondrial dysfunction and tubular damage in diabetic nephropathy. It was found that TIPE1 disrupted mitochondrial homeostasis in renal tubular epithelial cells (RTECs) by destabilizing the mitophagy receptor PHB2, leading to tubular damage and progression of diabetic nephropathy. This suggests that TIPE1 could serve as a potential therapeutic target for preventing diabetic nephropathy progression.

JAML promotes acute kidney injury mainly through a macrophage-dependent mechanism

Wei Huang et al.

Summary: This study elucidated the role of JAML in the pathogenesis of acute kidney injury (AKI). It was found that JAML promotes AKI mainly through a macrophage-dependent mechanism, and JAML-mediated macrophage phenotype polarization and efferocytosis are critical signal transduction pathways linking inflammatory responses to AKI. JAML may regulate macrophages through a macrophage-inducible C-type lectin-dependent mechanism.

Comprehensive identification of immuno-related transcriptional signature for active pulmonary tuberculosis by integrated analysis of array and single cell RNA-seq

Yuzhong Xu et al.

Summary: Through genomic technology, we found that immune-related transcriptional features can effectively differentiate tuberculosis, latent tuberculosis infection, and healthy controls. Key immune-related genes ADM, IFIT3, and SERPING1 are closely associated with TB, and ADM expression significantly increases in TB patients, with elevated levels in TB myeloid cells. These results provide useful and novel insights into exploring potential candidate genes for TB diagnosis and intervention.

Summary: In this study, IDO1 was identified as a key immune gene in diabetic nephropathy (DN), with its expression correlated with specific immune cell types. It serves as a diagnostic and prognostic biomarker for DN, playing a vital role in immune cell infiltration in DN as determined using microarray data and CIBERSORTx for the first time.

The Attenuation of Diabetic Nephropathy by Annexin A1 via Regulation of Lipid Metabolism Through the AMPK/PPARα/CPT1b Pathway

Liang Wu et al.

Summary: ANXA1 plays a crucial role in the pathogenesis of diabetic nephropathy by regulating lipid metabolism and inflammation resolution. Its deficiency aggravates renal injuries, while ANXA1 mimetic peptide shows therapeutic effects against lipid toxicity in diabetic mice. In human PTECs, ANXA1 silencing leads to deleterious effects driven by FPR2/ALX, highlighting its potential as a therapeutic target for DN.

Summary: Higher plasma levels of KIM-1, TNFR-1, TNFR-2, MCP-1, suPAR, and YKL-40 were associated with increased risk of progression of diabetic kidney disease. TNFR-2 had the highest risk after accounting for other biomarkers. These findings validate previous literature on TNFR-1, TNFR-2, and KIM-1 and provide new insights into the influence of suPAR and YKL-40 as plasma biomarkers that require validation.
