In vitro assessment for cytotoxicity screening of new antimalarial candidates

Mariana Rodrigues Espindola et al.

Summary: In antimalarial research, there is a lack of standard procedures for assessing the toxicity of drug candidates. This study compared different in vitro cytotoxicity methods for evaluating the toxicity of antimalarial drugs, and found that the TOV-21G, HepG2 and WI-26VA4 cell lines were suitable for evaluating the cytotoxicity of potential bioactive compounds.

Indirect effects of the COVID-19 pandemic on malaria intervention coverage, morbidity, and mortality in Africa: a geospatial modelling analysis

Daniel J. Weiss et al.

Summary: This study evaluated the impact of disruptions to malaria control on malaria incidence and mortality in Africa. Findings show that reductions in access to effective antimalarial drug treatment and insecticide-treated net coverage could lead to increased malaria burden, potentially doubling malaria mortality in 2020.

Identification and Assessment of Plasmodium berghei Merozoites and Cell Cycle by Flow Cytometry

Qigui Li et al.

Summary: Flow cytometry (FCM) allows reliable identification and quantification of blood stage parasites in each individual life stage, including merozoites, offering a faster alternative to microscopy. This integrated whole life stage model, particularly with confirmed determination of merozoite population, holds promise for drug and vaccine research in malaria therapy and prophylaxis.

The MTT Assay: Utility, Limitations, Pitfalls, and Interpretation in Bulk and Single-Cell Analysis

Mahshid Ghasemi et al.

Summary: The MTT assay is commonly used in studies of cell toxicity, but it is often misinterpreted. Our research found that the assay measurements are influenced by various factors and require optimization and rational interpretation of the data to avoid misleading conclusions. While the assay can be a powerful tool, it also has limitations.

Natural Products from Actinomycetes Associated with Marine Organisms

Jianing Chen et al.

Summary: Actinomycetes are a rich source of bioactive secondary metabolites and play a critical role in pharmaceutical research, especially those associated with marine organisms. Streptomyces, Micromonospora, and Nocardiopsis are dominant producers of secondary metabolites, with alkaloids, polyketides, and peptides being the major classes of natural products, mostly with antimicrobial and cytotoxic activity. Further attention should be paid to actinomycetes with multiple host organisms due to their special ecological status and genetic factors.

Endah Setyaningrum et al.

Summary: The study assessed the ability of unpurified fermentation metabolites from Streptomyces to inhibit Plasmodium parasites, with the extract showing the presence of saponins and triterpenoids and yielding an IC50 value of 11.07 g/mL. Further examinations of this extract, including assessment of off-target effects, are warranted.
