Summary: The study found that GeneXpert assay is reproducible with less than 2% blood contamination, but unreliable beyond 5% and completely inhibits PCR at 20% blood contamination. Boiling at 95 degrees C for 5 minutes can recover previously undetectable levels of MTB DNA.

Global Tuberculosis Report 2020-Reflections on the Global TB burden, treatment and prevention efforts

Summary: The global TB report for October 2020 highlights slow progress in TB control efforts and the need to ramp up screening and diagnostic test services. Key drivers of TB, including undernutrition, poverty, diabetes, tobacco smoking, and household air pollution, must be addressed to achieve WHO 2035 TB care and prevention targets. Global focus is needed on the development of new vaccines for TB using new technological methods.

Summary: Xpert Ultra and Xpert MTB/RIF are effective tools for diagnosing extrapulmonary tuberculosis, particularly for detecting rifampicin resistance cases. Xpert Ultra has higher sensitivity and lower specificity compared to Xpert MTB/RIF in diagnosing tuberculous meningitis, while both tests have similar sensitivity and specificity in detecting rifampicin resistance. Future research should address the limitations associated with using culture as a reference standard in paucibacillary specimens.
